However, the advent of this new technology restricted its use, almost exclusively, to research laboratories and specialized centers in molecular biology, due to its dependence of expensive and specific devices and the complexity of procedures. As knowledge advanced and new technologies emerged, the limitations of the PCR technique were also targets for improvement. In this way, since 1990s, few years after the PCR technique advent, new approaches were created, allowing the execution of the reaction under isothermal conditions and demanding less or none specific devices. Since 1990s, several isothermal amplification techniques have been created and applied in diagnosis of infectious diseases, such as: NASBA™ (Nucleic Acid Sequence-Based Amplification), 3SR (Self-Sustained Sequence Replication), SDA (Strand Displacement Amplification), RCA (Rolling Circle Amplification), HDA (Helicase-Dependent Amplification), CPA (Cross Priming Amplification), PSR (Polymerase Spiral Reaction) and LAMP (LoopMediated Isothermal Amplification). These techiniques have created a range of alternatives and new concepts of nucleic acids amplification.
