This study aimed to establish the organotypic tissue culture protocol (CTO) for colorectal cancer tumor tissue (CCR) derived from lesions removed in surgery from a patient at LIGA Norte-Rio-grandense Contra o Cancer, Natal, RN. The production of cuts in the biopsied tissue of the mentioned lesions for the development of CTO came from a patient with CCR without previous treatment. The ex vivo slices were cultivated under specific conditions in order to keep them for 4 hours and 24 hours. Some tumor sections were submitted to treatments with oxaliplatin (OXA) at different concentrations and others had contact only with DMEM. At the end of these times, with the purpose of verifying the tissue viability of the CTO, a double-blind histopathological analysis of parameters correlated to the composition of the tumor microenvironment (TM) was performed, as well as the investigation of the expression of TGF-b, a predictive marker of MT immunosuppression. The general morphology of the organotypic control tissues was maintained after 4 hours and after 24 hours there were modifications and rearrangements in the microstructure of the tumor. An increase in necrosis was observed over the time of the experiment, as well as an attenuation of the population of immune cells and glands. Furthermore, in the treatment with the lowest concentration of OXA, the tumor seemed to restructure with more presence of fibroblastic tissue. TGF-β marked more well-preserved tissues, especially in CTOs that had transformed intestinal glands. According to these results, it can be concluded that the technique maintains tissue viability over time, with some conformational changes arising from environmental changes, as well as the limitations of the technique.
