Low level laser therapy (LLLT) has been suggested as a tool to promote bio-stimulation of various cell types in vitro. This study aimed to verify the efficacy of LLLT in the proliferation of preosteoblast cells (MC3T3-E1) cultured on PLA film. We evaluated three groups: I (control) – cells cultured on plastic surface; II (PLA) – cells cultured on PLA film; and III (PLA + Laser) – cells cultured on PLA film and submitted to irradiation with diode laser (InGaAIP) by using a wavelength of 830 nm, output power 30 mW, and a single dose of 4 J/cm2, with the radiation emitting continuous radiation. The experiment was conducted in quadruplicate at four time intervals: 6, 24, 48, and 72 h. Cell adhesion was analyzed 6 h after the cell culture by counting cells stained by toluidine blue. Cell viability and proliferation were evaluated by counting the cells in all three groups using trypan blue exclusion assay (24, 48, and 72 h) and photomicrographs of the biomaterial/plastic interface (6, 24, 48, and 72 h). The results showed that group III exhibited a higher proliferation compared to the groups I and II.
