Análise de enriquecimento de genes obtidos por RNA-seq de infectados por Vírus Zika e Chikungunya: uma abordagem acerca da assinatura biológica no hospedeiro. 2019. 50f. Trabalho de Conclusão de Curso (Graduação em Farmácia) - Departamento de Farmácia, Universidade Federal do Rio Grande do Norte, Natal, 2019.https://repositorio.ufrn.br/handle/123456789/35773Currently, several computational tools have been used in biomedical analysis of genomic data from complex and infectious diseases, aiming at increasing computational speed and performance, as well as analytical accuracy, aiming at clinical resolution. In this regard, recent outbreaks of Zika virus infection (ZIKV) have highlighted the importance of recognizing this syndrome as an acute public health case, and its study is of primary importance for this purpose. Thus, ZIKV-infected data were used during the 2015 outbreak; Guillain-Barre syndrome, whose inflammatory process in peripheral nerves may be caused by viral infections, including ZIKV. Analyzes were based on data from High Throughput Sequencing, in which transcriptomics using NGS (RNA-seq) were used in individuals recovered from ZIKV infection, those who developed Guillain-Barré syndrome due to infection with ZIKV, besides being infected by chikungunya virus (CHIKV). In order to identify biological signatures in pathways, genes and biological processes, in addition to differentiating host phenotypes under the influence of these different infections, the Gene Set Enrichment Analysis (GSEA) functional gene enrichment methodology was used. The results indicate that developmental pathways (eye morphogenesis, ciliary movement), inflammation-related pathways and innate immune system, as well as synaptic transmission pathways, are upregulated recovered from GBS. The same is indicated for recovered by ZIKV, 400 days after infection. In GBS patients, cell cycle pathways and neurogenesis can be observed as overregulated, with mitophagy and downregulated neuronal maturation, indicating that GBS recovered individuals tend to recover normal functions by decreasing gene expression in these pathways. Regarding CHIKV, pathways linked to innate immune system regulation (mainly linked to viral RNA recognition), cell cycle, host defense responses etc are overregulated. Those pathways linked to biosynthetic pathways, ribosome production, and cellular signaling during inflammatory processes are downregulated, indicating cellular processes that lead to normality and correction of modification processes during infection by these viruses.
