The regulation of serotonin levels during development is extremely important for the characteristic development of the brain. Changes in serotonergic signaling, both at low and high levels, appear to be harmful, leading to several behavioral deficits. Early exposure to antidepressants, such as fluoxetine, during pregnancy or lactation leads to several deleterious effects on the anatomical and functional organization in different brain circuits, including those related to the circadian rhythm. In recent years, different authors have investigated these harmful effects and the mechanisms by which they lead to structural and functional disorganization in specific neural networks, as well as altered behavioral expression in the individual. However, there is a lack of work at the neurochemical level on structures of the circadian timing system. In this context, this work aims to investigate whether early hyperstimulation of the serotonergic system, through perinatal exposure to an SSRI, can affect the circadian rhythm of activity and rest, especially investigating the expression of neurotransmitters associated with the circadian rhythm at different stages of the offspring's life ( P15, P45 and P90). In this study, the rhythm of locomotor activity-rest will be tested, and the visualization of chemical oscillatory markers of the circadian system (NPY, 5-HT and BMAL1) in two zeitgebers (ZT3 and ZT14) in male and female rats exposed early (period of development GD13-GD21) fluoxetine (20 mg/kg). For this, 20 females and 10 males were used for mating. With the adult offspring, we performed daily recordings of the locomotor activity-rest rhythm using activity sensors attached to individual cages over the course of four weeks, two in light- dark and two in constant darkness. Some animals were perfused at three stages of life (P15, P45 and P90) to identify the expression of neurochemical markers in the suprachiasmatic nucleus, the intergeniculate leaflet (FIG) and the raphe nuclei. Changes were seen in the behavioral variables of the activity-rest rhythm in the fluoxetine group, as well as changes in the expression of markers used at work, in the fluoxetine and control groups, of males and females.pt-BRAcesso AbertoSerotoninaRitmo CircadianoFluoxetinaImplicações do tratamento perinatal da fluoxetina no ritmo circadiano de atividade- repouso e em componentes do sistema de temporização de ratos wistar, machos e fêmeasdoctoralThesisCIENCIAS BIOLOGICAS
