So, the present study aimed to analyze in which stage or stages of this treatment the color change occurs. It is an experimental in vitro study where 75 bovine incisors (n=15), divided in 5 groups: GP- complete protocol, GA- triple antibiotic paste, GS- blood, GW- WMTA, GC- serum (control). The color evaluation was performed with the Easyshade® device, with the CIE-L*a*b* parameters. And the readings were performed in triplicate for each group, before and after the first session, after 24, 48 and 72 hours, one and three months after the protocol exexution. The Statistical analysis was performed using the split-plot analysis of variance test, and the average value in all groups was adjusted by Bonferroni, with a significance level of 95%. For intergroup and intragroup analysis, the statistical comparisons were made through the ANOVA test with the Tukey Post Hoc. According to the multivariate comparison, considering the total average of the degree of darkening, the blood and protocol groups presented more significant darkening (p<0.05). In the intergroup analysis, the group containing blood presented a higher darkening degree from 15 days to three months (p<0,05) and the protocol group presented coronary discoloration on the first and third months in relation to the other groups (p<0,05). In the intragroup analysis, the antibiotic and WMTA groups presented a decrease on the darkening in 15 days compared to the baseline (p<0,05) and then remained stable, the group containing blood had an increase after 15 days (p<0,05) and the protocol group, in 15 days showed coronary discoloration after the introduction of blood on the root canal, showing variation to the baseline on the third month (p<0,01).
