Caracterização fitoquímica e avaliação do efeito antioxidante e citotóxico de extratos das folhas de imburana de espinho (Commiphora leptophloeos) (Mart.) J.B. Gillett (Burseraceae) . 2018. 81f. Dissertação (Mestrado em Bioquímica) - Centro de Biociências, Universidade Federal do Rio Grande do Norte, Natal, 2018.https://repositorio.ufrn.br/jspui/handle/123456789/25992Commiphora leptophloeos (Mart.) J. B. Gillett, folk knwon as “Imburana de espinho”, is a native plant from Caating. This plant is popular used for diferent patologies. However, few scientific data is available related to its biological activities. In this work, extracts from fresh leaves through a serial extraction using different solvents in growing polarity order were obtained: hexanic (EH), chloroformic (CE) ethanolic (EE), methanolic (EM), residual aqueous (EAR). Besides, it was done another extract called as water extract (EA) similar as the one produced in folk medicine. The phytochemical characterization was done by measuring the phenolic compounds, total proteins and sugars. It was also done a qualitative assay by thinlayer chromatography (TLC) and by ultra-high performance liquid chromatography (UPLC). Furthermore, it was also analyzed in vitro antioxidant activity and in vitro cell viability by MTT assay. The results obtained with TLC showed the presence of phenolic compounds, terpenes, flavonoids, phenolic acids, tannins and saponins in special for the EE and EM. The Co-TLC analysis showed the presence of flavonoids luteolin and isoquercetina for the EE and EM. Moreover, UPLC analysis identified rutin in these samples. The antioxidant assays showed that all extracts have the antioxidant capacity in vitro, in special for the EE and EM. Based on these results, it was analyzed the EE effect on the animal model Caenorhabditis elegans. This in vivo assay showed that the EE was able to reduce in 50% of the ROS intracellular levels from this animal model. Furthermore, the MTT assay showed that EE was not cytotoxicfor 3T3 cell lineage; on the other hand, for the B16-F10 cell lineage (tumor cell) it was observed a 45% reduction. Taken together, these results propose that C. leptophloeos has a potential source of bioactive compounds with antioxidant and antitumor potential.
