The present work consists on a systematic review about the pharmacokinetics of phenytoin into biological matrices. A search in Pubmed database was carried out using “phenytoin”, “pharmacokinetic” and “chromatography” as keywords. Articles avaiable in a free complete version and published from 1995 to 2006 were selected. Studies which were carried out exclusively in animals or those which performed just in vitro assays were excluded. The following parameters were considered: author(s), year of publication, aims, methodology and conclusion. Sixten articles were reported. It was possible to verify that pharmacokinetic of phenytoin has been studied in different biological matrices using suitable spectroscopic methods for its detection. This systematic review proved that High Performance Liquid Chromatography was the most used method and also that there is a great variability of studies developed with phenytoin. This way, many studies have been carried out in order to improve the methods for extraction and detection of this molecule in different biological matrices. It is known that low concentrations of phenytoin in maternal milk, umbilical cord, plasma and hair limit the detection of this drug.
