Estabelecimento da carcinomatose peritoneal como modelo do câncer colorretal metastático para testes de fármacos e nanosistemas. 2022. 49 f. Trabalho de Conclusão de Curso (Graduação em Biomedicina) – Universidade Federal do Rio Grande do Norte, Natal, 2022.https://repositorio.ufrn.br/handle/123456789/51018Peritoneal carcinomatosis (PC) is one of the forms of metastasis that most increases the mortality of colorectal cancer (CRC). Therefore, the need for a previously established animal model is extremely relevant to start studies with experimental treatments. In this sense, the objective of this work was to establish the model of peritoneal carcinomatosis to be used as a study tool in testing drugs, compounds and nanosystems. For this, five animals were submitted, each one, to the inoculation of 1x10⁶ CT26 cells in the peritoneum. Twelve days after inoculation, euthanasia and weight assessments were performed, as well as sample collections for hematological, histopathological, immunohistochemical and immunofluorescence analyses. Although there was a decrease in weight and red blood cells in the CP model animals, the result was not statistically significant. Likewise, there was visible leukopenia, but this was not considered statistically significant (p>0.05). In the lungs (p<0.05), liver (p<0.05) and spleen (p<0.05), relevant metastatic niches were found with increased expression of the tumor marker CD26 in metastatic organs, such as the liver (p<0. 05) and lung (p<0.05), whereas CD163 expression increased in liver (p<0.05) and peritoneal tumor (p<0.05). On the other hand, CD86 expression increased only in the lung (p<0.05). It is clear, therefore, that the CP study model is an important tool for studying signaling pathways in metastases of aggressive RCC cancers, and more importantly, blocking these pathways with tests of new compounds, drugs and nanosystems.Attribution-NonCommercial-NoDerivs 3.0 Brazilhttp://creativecommons.org/licenses/by-nc-nd/3.0/br/Cancer colorretalCarcinomatose peritonealMetástaseProtocoloEstabelecimento da carcinomatose peritoneal como modelo do câncer colorretal metastático para testes de fármacos e nanosistemasEstablishment of peritoneal carcinomatosis as a model of metastatic colorectal cancer for drug and nanosystem testingbachelorThesisCNPQ::CIENCIAS BIOLOGICAS
