The use of the matrix systems based on polymers is an interesting alternative for prolonged the release of drug administered by oral route. The present work has the aim to development a matrix system based on the hydrophilic polymer hydroxypropilmetylcelullose (HPMC) for prolonged the release of triamcinolone and additionally to study the kinetics mechanism of in vitro. Thus, it was prepared four formulations based on microcrystalline cellulose, magnesium stearate and HPMC at different concentrations (30, 40, 50 and 60%w/w)and for every formulation were obtained three different hardness (5.0; 7.5 and 10.0KgF). The pharmacopoeial quality control tests were conducted and all formulations were approved. In the swelling test, the formulation with hardness of 7.5 KgF showed higher swelling percent and it was selected to incorporate triamcinolone (10mg). The analytical methodology used was validated according to parameters established by Brazilian Regulatory Agency (ANVISA). The mean concentration drug was 94.77%.
