Mapeamento de epítopos imunogênicos de células T de Leishmania infantum para vacinas contra Leishmaniose visceral. 2021. 50 f. Monografia (Graduação em Biomedicina) – Universidade Federal do Rio Grande do Norte, Natal, 2021.https://repositorio.ufrn.br/handle/123456789/44996Leishmania infantum is the main species responsible for human visceral leishmaniasis (HVL) disease in Brazil. The parasite is transmitted by the bite of a sandfly Lutzomya longipalpis during a blood meal, showing domestics dogs as the main reservoirs. The difficulty of developing a vaccine makes this type of prospective study extremely important, since cases of resistance to antimonials have been reported, increasing the need for prophylactic measures. Advances in bioinformatics tools allow the possibility of exploring specific immunogenic antigens without spending resources in vitro that would otherwise be expensive. This study was carried out to screen for immunodominant T cell epitopes from Leishmania infantum chagasi contained in the amino acid sequences of antigens 314, 319, 419, 425, 503 and 648 obtained in previous studies of our group as a target for vaccine development. Using the TepiTool program, we found potential T cell epitopes for each sequence analyzed and, among these, only the non-allergenic epitopes described by AllerTop were selected. The predominant HLA alleles were selected for each class, described by the IEDB as predominant in the world population. The selected epitopes were also evaluated in silico to produce cytokines such as IL-4 and IFN- ℽ. Homologies were found for all Leishmania sequences. Altogether, 35 immunogenic epitopes were generated capable of generating an immune response and, among these, 28 were able to induce cytokine production.
