Universidade Federal do Rio Grande do Norte, 2019.https://repositorio.ufrn.br/handle/123456789/47308Agomelatine, an unconventional antidepressant used in clinical practice, is a compound that acts as a melatonin receptor agonist and a serotonergic antagonist, it has been pointed out in clinical and preclinical studies as a new alternative for treating this condition. However, there are still few studies with females, that according to some studies, are affected twice as much as males when it comes to depressive disorder. The aim of the present study was to evaluate whether the administration of agomelatine alters depression-related behavioral responses in rats undergoing forced swimming test. For this purpose we used female Wistar rats, approximately 90 days old, which were submitted to agomelatine administration (50 mg / kg/ gavage) dissolved in saline solution (0.9% NaCl) for 24 days. The estrous cycle of the rats was followed through the maginal swab, for the evaluation of the locomotor activity the open field test was used and the evaluation of the depressive behaviors was performed through the forced swimming test. The results show that there was no significant change in female locomotion and that the 50 mg / kg dose significantly altered the forced swim test parameters increasing the immobility and decreasing the climbing results that suggests a depressive type effect.
