A total of 40 samples were analyzed, equally divided between the two methods, to determine serum hemoglobin levels and the degree of hemolysis. The samples were processed following a standardized protocol, including centrifugation at 3,500 rpm for 10 minutes, and hemoglobin levels were quantified. The results showed that the vacuum system significantly reduced hemolysis levels, with 65% of the samples classified as GH1 (low grade) and 25% as GH2 (moderate grade), while the conventional method showed a higher frequency of GH3 (20%) and GH4 (10%) grades. The conventional group had a mean serum hemoglobin level of 76.8 ± 9.3 mg/dL, while the vacuum group had 49.0 ± 6.4 mg/dL (p = 0.02, unpaired t-test). Consequently, samples collected using the vacuum system demonstrated higher quality and less analytical interference, which is crucial for sensitive biochemical analyses such as enzyme and electrolyte measurements. These findings indicate that the vacuum system is more effective in preserving cell integrity and sample quality, aligning with the principles of the 3Rs (replacement, reduction, and refinement).
