Estudo de polimorfismos na região promotora do gene IL-10 em mulheres com lesão intraepitelial e cancêr cervical. 2019. 48f. Trabalho de Conclusão de Curso (Graduação em Biomedicina) - Centro de Biociências, Universidade Federal do Rio Grande do Norte, Natal, 2019.https://repositorio.ufrn.br/handle/123456789/43201In Brazil, cervical cancer occupies the third position in the ranking of cancer incidence in the female population. Human papillomavirus (HPV) is the main causative agent of cervical cancer and precursor lesions, and it is important to note that HPV often uses strategies to evade the immune response using mechanisms that involve immunoregulation. In the tumor microenvironment, some immunoregulatory molecules have been highlighted and they have contributed to the progression of cervical cancer. IL-10 is an immunosuppressive cytokine, often elevated in the tumor microenvironment. The gene encoding human IL-10 is located on chromosome 1 (1q31-32). Single-base polymorphisms (SNPs) have also been identified in the IL-10 promoter region that may influence cytokine production, and therefore play a role in cancer susceptibility. In this study the SNPs -1082A> G ( rs1800896 ) and -819C> T ( rs1800871 ) of the IL-10 gene were analyzed from peripheral blood samples from 317 Brazilian women, 142 women with alteration in the cervix (altered group), framed with SIL squamous intraepithelial lesion (SIL) (n = 101, 24 patients with low grade lesion and 77 with high grade lesion) and cervical cancer (CC) (n = 41) and 167 representing healthy women. The objective was to associate the allelic and genotypic frequency of IL-10 SNPs and to associate with the development of SIL and CC. As a result, in polymorphism rs1800896 , the GG genotype frequency was statistically lower in the altered group (SIL and CC) and in the SIL group, compared to the control group (OR = 0.35, 95% CI = 0.14-0 , 88, p = 0.026, OR = 0.36, 95% CI = 0.13-1.00, p = 0.049, respectively). The CC group, compared to the control, did not show any significant difference, nor was there a statistical difference between the cases and control in the polysorphism rs1800871 (p> 0.05). Thus, it can be concluded that, in the polymorphism rs1800896 , the GG genotype was not associated with the development of SIL and cervical cancer.
