Tocilizumab (TCZ) is an IL-6 receptor monoclonal antibody, and its systemic administration has an anti-inflammatory effect on this condition. The aim of the present study was to evaluate the effect of local application of TCZ in an experimental model of periodontitis. 50 Wistar male rats were randomly divided into three groups: CG- control (n=10), EP- experimental periodontitis (n=20), and EP + TCZ- experimental periodontitis + TCZ 2mg/kg (n=20). On day 0, the animals received local application of 10µL of saline solution (CG and EP groups) or TCZ (EP + TCZ group) in each palatine papilla of the upper left second molar (ULSM). On the third experimental day, animals from the EP and EP + TCZ groups underwent periodontitis induction by placing a 3.0 nylon thread around the ULSM. On the 14th day, the animals were euthanized, and blood, maxillae and gingival samples were removed for biochemical (liver and kidney markers), microtomography, histologic, cytokine assay by ELISA (IL-6) and RT-PCR analysis (SOD and GPx). Data obtained were evaluated for normality and submitted to ANOVA or Kruskall-Wallis tests. TCZ did not change serum levels of liver and kidney markers, and was able to attenuate linear alveolar bone loss, preserve the number of bone trabeculae, and reduce the severity of inflammation and gingival tissue levels of IL-6 (p<0.05).
