N,N-dimethyltryptamine (N,N-DMT) is an indolic alkaloid studied for its potential in the treatment of depression, anxiety disorders and addiction. Therefore, the aim of this study was to obtain N,N-DMT through extraction from Mimosa tenuiflora (known as Jurema-Preta) and synthesis routes. The substance extracted by the standard method showed an average purity of 99.0 ± 0.4%, making it viable for clinical and preclinical trials. Despite their therapeutic potential, many extraction methods have not been consolidated or optimized. Thus, in order to optimize the method employed, a fractional factorial design (26-1) was used with variations in the parameters of particle size, concentration of the alkaline solution, mixing method, temperature, mixing time and eluent-solid ratio. All these parameters, except the mixing method, proved to be influential. The optimized method, using an eluent-to-solid ratio of 50 mL/g, a temperature of 55 ºC, a 2.0 M NaOH solution, a particle size of 75 μm and 8 hours of mixing, resulted in a 3.5-fold increase in yield compared to the standard method, without compromising the purity of the final product (98.5%). The synthesis of N,N-DMT gave a yield of 52.1%, but required additional analytical techniques to better characterize the product. A comparative analysis of production costs was also carried out, evaluating the methods of synthesis, standard extraction, optimized extraction and a hybrid method. The latter, which combines parameters from the standard and optimized methods, proved to be more cost-effective.
