These structural modifications aimed to enhance its physicochemical properties and broaden its antimicrobial activity, making it a promising tool in combating resistant infections. This study developed a robust and efficient analytical method for its analysis using the Analytical Quality by Design (AQbD) approach in conjunction with the UHPLC/MS-ESI TOF technique. A full factorial experimental design was applied to evaluate the impact of critical variables such as column length, mobile phase composition, and flow rate. Methods 9, 18, and 27 yielded promising results, all using a 150 mm column and a mobile phase composition of 50%. The differentiating variable among these methods was the flow rate, which ranged from 0.1, 0.2, to 0.3 mL/min, respectively. Pareto and response surface graphs highlighted the relevance of the mobile phase and column for optimizing chromatographic efficiency, retention time, and peak symmetry, whereas the flow had a lesser statistical impact. This study significantly contributes to the analysis of bioactive peptides, offering a practical and innovative tool for quality control in pharmaceutical and biotechnological laboratories.
