Diet plays a central role in modulating inflammation, oxidative stress, and the clinical progression of the disease. Although dietary parameters such as the Dietary Antioxidant Quality Score (DAQS), the Dietary Total Antioxidant Capacity (DTAC), and the NOVA classification are used to assess diet quality, studies exploring their relationship with clinical outcomes in individuals with ALS remain scarce. This study aimed to evaluate the relationship between the dietary parameters DAQS and DTAC and clinical variables in individuals with ALS. A descriptive cross-sectional study was conducted with 68 individuals followed at the multidisciplinary ALS outpatient clinic of the Onofre Lopes University Hospital – HUOL (UFRN). The sample was characterized according to demographic, clinical, and nutritional characteristics. ALS progression and severity were assessed using the revised ALS Functional Rating Scale (ALSFRS-R). Nutritional status was determined by Body Mass Index [BMI (kg/m²)], categorized as underweight, normal weight, and overweight. Dietary intake was assessed using two nonconsecutive 24-hour dietary recalls (24HRs). Energy and micronutrient intake were analyzed using the Virtual Nutri Plus® software. The DAQS was calculated based on the intake of vitamin A, vitamin C, vitamin E, zinc, and selenium, and categorized as “low/medium” or “high.” The DTAC, expressed in mmol/1000 kcal/day, was estimated using literature data based on the Ferric Reducing Antioxidant Power (FRAP) method, and classified as “high” or “low” according to the median value. DTAC data were also described as the percentage contribution of each NOVA food classification group (unprocessed and minimally processed foods, processed culinary ingredients, processed foods, and ultra-processed foods) to the total DTAC. All collected variables underwent descriptive analysis. Variables with normal distribution were analyzed using the independent t-test, while non-normally distributed variables were analyzed using the non-parametric Mann–Whitney test. These tests were used to compare DAQS (high vs. low/medium) and DTAC (high vs. low) groups to demographic and clinical characteristics. Effect sizes were calculated for all statistical tests. Analyses were performed using R software, version 4.5.0, with a significance level of 5% (α = 0.05). Participants differed significantly by sex, with males presenting higher ALSFRS-R scores (p = 0.013), energy intake (kcal/day, p < 0.001), vitamin C intake (mg/day, p = 0.004), and zinc intake (mg/day, p = 0.018). The majority of DTAC (79.8%) originated from Group 1 foods according to the NOVA classification. Participants with a high DAQS had a shorter symptom duration in months (p = 0.023) and higher ALSFRS-R scores (p = 0.047) compared to those with low/medium DAQS. Higher BMI values were observed in the high DTAC group, differing significantly from the low DTAC group (p = 0.039). In conclusion, both DAQS and DTAC were associated with relevant clinical indicators in individuals with ALS, including nutritional status, functional capacity, and disease progression. The Nutrition Care Process (NCP) is a systematic and standardized care management model that consists of four steps to address nutritional problems and provide effective, high-quality care to individuals. To document nutrition care, a standardized language is used for each step, known as the Nutrition Care Process Terminology (NCPT). To assess the implementation of the NCP/NCPT, audit instruments are employed, with the NCP-Quest being the most up-to-date and validated tool in the United States. In 2020, in Brazil, the "Consórcio para Pesquisa e Implementação do PCN no Brasil" was established, marking the beginning of an effort to implement the NCP/NCPT in the country. Thus, the objective of this study was to validate the NCP-Quest for Brazilian Portuguese. This is a methodological study, divided into two phases: (1) Validation of the translated content and (2) Assessment of the instrument's reliability. The validity of the translated content was analyzed using the Content Validity Index (CVI), both for individual items (I-CVI) and for the overall instrument (S-CVI). The instrument's reliability was assessed using α-Krippendorff’s, the percentage of agreement among raters, and the intraclass correlation coefficient (ICC) for each item, NCP section, all items combined, and the total score. The I-CVI ranged from 0.80 to 1.00, and the SCVI was 0.988 in the first round and 0.992 in the second round of translated content validation. Inter-rater reliability for the total score showed an ICC of 0.964 (95% CI = 0.850 – 0.992) for average measures and 0.930 (95% CI = 0.739 – 0.983) for single measures, demonstrating excellent agreement. α-Krippendorff’s was 0.66, indicating sufficient agreement. When evaluating individual items, some showed a negative α-Krippendorff’s, indicating a higher level of disagreement than expected by chance, which may be related to different interpretations and the subjectivity of certain instrument items. However, the impact of UPF consumption on the antioxidant profile of women during lactation is still unknown. Considering that breastfeeding is essential for the healthy development of the child, providing growth factors, nutrients and a powerful antioxidant capacity via breast milk, the present study aimed to investigate the relationship between UPF consumption and the antioxidant profile of breast milk and the diet of 136 lactating women treated in primary health care in the city of Natal-RN, with a median of 66 days postpartum. The study was approved by the Research Ethics Committee of HUOL/UFRN (Opinion No. 4,199,673). Socioeconomic and health information, anthropometric measurements, 24-hour recalls (R24h) adapted to the New Classification, and aliquots of breast milk from a single breast were collected. To determine antioxidant markers in breast milk, the enzymes superoxide dismutase (SOD) and catalase were analyzed using commercial kits by plate reading, as well as the quantification of alpha-tocopherol by ultra-highperformance liquid chromatography. The theoretical estimate of vitamin E supply via analyzed milk was also evaluated (4 mg of vitamin E in 780 mL per day). Food consumption was assessed according to the caloric participation of UPF in the diet (% EAUP) and the antioxidant profile of the diet according to the dietary intake of vitamin E (mg/day and mg/1000 kcal) and total antioxidant capacity of the diet (CATd mmol/day). Lactating women were grouped into tertiles of % EAUP. The association between kcal of UPF and markers of the antioxidant profile in milk and diet were analyzed by adjusted multiple linear regression models. AUP contributed 24.3% of the calories in the diet and it was observed that women in the highest tertile of %EAUP had a higher intake of proteins, carbohydrates, lipids and fibers and CATd index (p<0.05). In breast milk, the alpha-tocopherol concentration presented a median of 151.4 (Q1-Q3 101.0-258.0) μg/dL and 77.8% of the milk analyzed did not reach, theoretically, 4 mg/780 mL of vitamin E. The SOD enzyme obtained a mean of 0.2 (0.1) U/mg-1 and Catalase a median of 60.1 (Q1-Q3 10.6-60.1) nmol/mg-1, with a positive relationship being found between UPF consumption (kcal) and the concentration of SOD (β = 0.00 95% CI -0.00-0.00; p<0.007) and CATd (β = 0.01 95% CI 0.00-0.02 p<0.001). Dietary treatment requires lipid restriction, with a maximum of 10 to 15% of total daily energy intake or 15 to 20 g of total fat per day, which can impact the intake of essential micronutrients such as selenium, zinc, and iron. However, there is a scarcity of studies evaluating the nutritional profile of these micronutrients in patients with FCS. Therefore, the objective of this study was to evaluate the nutritional and biochemical profile of selenium, zinc, and iron in patients with FCS, considering dietary intake and serum concentrations of these micronutrients. This is a case-control study with 11 FCS patients and 11 matched controls. Clinical, anthropometric, socioeconomic, biochemical, and dietary intake aspects were assessed. The case group was stratified into homozygous and heterozygous, while the control group was divided, according to the matching criteria, into Control 1 and Control 2. Statistical analyses included the Kruskal-Wallis test and Fisher's exact test, with a significance level of p<0.05. A statistically significant difference was observed in the lipid profile, with higher concentrations of VLDL triglycerides and reduced HDL and Apolipoprotein A in the FCS group, particularly more severe in the homozygous group. Although no serum selenium, zinc, or iron deficiencies were identified, the data indicate a risk of nutritional inadequacy in the consultation function below the recommendations, especially in women. The results demonstrate changes in the lipid profile and potential risks of inadequate selenium, zinc, and iron intake in patients with FCS. This study evaluated menus from five commercial restaurants in Natal, Brazil, considering social, health, and environmental dimensions. In the social dimension, a sociodemographic questionnaire and the Brazilian Food Insecurity Scale (EBIA) were applied to 261 workers (67.9% men; mean age 32 years; 53.5% with completed high school). Among them, 70.2% were responsible for food purchasing and 70.6% for domestic meal preparation. Food insecurity affected 53.3% of workers and was associated with income (p=0.001), education (p=0.023), family composition (p=0.020), and municipality of residence (p=0.028). In the health dimension, the nutritional status of 438 workers was assessed, with 68.3% presenting overweight or obesity; obesity prevalence was 30.5% among men and 42.7% among women. Across 111 menus analyzed, caloric density was within the recommended range, but with high lipid and sodium content, reduced carbohydrates, and adequate protein. In the environmental dimension, 54.1% of foods were supplied locally, 43.2% from within the state, and 2.7% from other regions of Brazil. Menu composition included 59.8% fresh or minimally processed foods, 9.3% processed, 16.5% ultra-processed, and 24.7% culinary ingredients. Beef showed the highest environmental impact, followed by poultry, eggs, and pork, with weekly averages of carbon footprint 2.3 kgCO2, water footprint 1.94 L, and ecological footprint 12.5 kg/m². Data triangulation, inspired by mixed-methods approaches with interpretation grids, was also adopted by crossing three axes: (i) macronutrients × obesity; (ii) caloric-nutritional adequacy × obesity × food insecurity; and (iii) proteins × carbon footprint × water footprint, in order to integrate results, minimize bias, and enhance complementarity of findings. Triangulation analysis showed that although menus had relatively similar macronutrient composition, obesity outcomes differed across restaurants, indicating a stronger influence of social and contextual determinants. Furthermore, caloric-nutritional adequacy and food insecurity alone did not explain these variations, reinforcing the need for integrated approaches. From an environmental perspective, higher carbon and water footprints were associated with menus containing more animal protein, with relevant differences depending on the type of protein offered. The findings reveal a concerning scenario: food service workers, including those employed in renowned restaurants, face food insecurity, highlighting social vulnerabilities and invisibility in protection policies. Ensuring dignity requires better working and dietary conditions. While menus met caloric requirements, the excess of lipids and sodium increases the risk of chronic diseases such as obesity. Considering that alternatives for the control of T2DM are necessary, inhibitors of enzymes involved in the process of carbohydrate digestion may be a possibility for glycemic control in humans. The present study aimed to evaluate the effect of the trypsin inhibitor isolated from tamarind seeds (Tamarindus indica L.) (ITT) and its peptides on α-amylase. After confirmation of the obtaining and characterization of the ITT, the analysis of its in vitro inhibitory activity against α-amylase was performed. Then, the interaction of the theoretical ITT (ITTp 56/287) and five of its derived peptides with α-amylase, as well as their functional properties, was also evaluated in silico, through Docking and Molecular Dynamics. The Potential Energy of Interaction (EPI), the Free Energy of Binding (ΔGbinding) were also calculated and the main electrostatic, hydrophobic and hydrogen bond interactions of the peptide-αamylase complex were described by means of three-dimensional representations (sticks). As a result, ITT presented 100% antitryptic activity and a molecular mass of approximately 21 kDa. The results of in vitro inhibition of α-amylase showed an inhibitory activity higher than 37%. These results were corroborated by computational analyses, which showed the strong interaction of the ITTp 56/287 complex and its peptides with the enzyme. The analyses of the Root Mean Square Deviation (RMSD) (up to 0.7 nm) and the Root Mean Square Fluctuation (RMSF) (up to 0.4 nm) showed low flexibility of the tested peptides, i.e., good stability. The EPI analysis showed - 705.08 kJ/mol for DTVHDTDGQVPL and -584.11 kJ/mol for TIAPACAPKPAR and the Free Energy of Binding (ΔGbinding) of -308.44 kJ/mol and -245.22 kJ/mol, respectively, confirming the previous results. The peptide sticks highlighted the electrostatic interactions, especially salt bridge, between the main residues that interacted in the complex (DTVHDTDGQVPL, TIAPACAPKPAR e TVSQTPIDIPIGLPVR). In addition, the bioactive potential predicted two candidates with good stability, high half-life and bioactivity in an intestinal simulation environment. The state of Rio Grande do Norte produces mainly extractive products for domestic consumption and use by the food industry. In this sense, it is important to promote the valorization of by-products generated by the processing of fruits, aiming at environmental, social, and economic sustainability. Therefore, the study's objective was to investigate the nutritional and technological properties of the flour obtained from the umbu peel, aiming at its use as an ingredient in bread production. The umbu peels were dried in a ventilated oven (55 °C for 24 h), followed by grinding to obtain the flour. The flour obtained was characterized according to morphology, granulometry, physical-chemical parameters, centesimal composition, and technological properties. Subsequently, four bread formulations were developed to partially replace wheat flour with umbu peel flour (standard – without umbu peel flour, and containing 5%, 10%, and 15% umbu peel flour). The breads were evaluated for centesimal composition and sensory attributes through acceptability tests, ideal scale, and purchase intention. Umbu peel flour presented heterogeneous particles, with variation in size and shape, demonstrating characteristics of integrality and presence of fibers, and classification as fine-granule flour according to the granulometry analysis. Regarding the physicochemical aspects and centesimal composition, the low pH (2.68 ± 0.00), high acidity (9.27 ± 0.07 g/100 g), and high total dietary fiber content (40.45 ± 0.065 g/100 g) stand out. The technological potential was evidenced by the high-water absorption capacity (5.08 g/g) and emulsifying stability (100%). Regarding the prepared bread, it was observed that the bread with 15% umbu peel flour had a higher ash content (2.72 ± 0.02%), protein (17.90 ± 0.50%), and fiber (3.13%). Regarding the sensory evaluation, the acceptance of the samples decreased with the increase in the concentration of umbu flour replacing wheat flour, with acceptability values of 83.8% for the standard bread, 72.8% for the 5% formulation, 57.3% for the 10% formulation, and 46.3% for the 15% formulation. The ideal scale indicated that attributes such as acidity, sweetness, and texture were the main factors that penalized the breads. The attitude test revealed that the formulation with 5% umbu flour had positive purchase intentions. During the pandemic, Brazil adopted social distancing measures to reduce the spread of the new virus. This context triggered changes in the population's eating practices, both positive and negative. In Brazil, the Food Guide for the Brazilian Population is the document that guides dietary practices, which can influence the perception of quality of life (QoL). This work includes two studies: cross-sectional and a cohort, and aims to identify adherence to the Food Guide and its association with the quality of life of adults and the elderly, before and after the COVID-19 pandemic. In both studies, the population consisted of adult and elderly residents of the city of Natal-RN and data were collected using a questionnaire on the Epicollect platform5, in the period 2019-2020 in the cross-sectional study and 2023-2024 in the cohort, containing sociodemographic, socioeconomic, anthropometric nutritional status, eating practices, QOL, and self-reported COVID-19 variables in the cohort. A multidimensional scale was used to analyze adherence to the Food Guide, resulting in the classifications “low”, ‘medium’ and “high”. QoL was analyzed using the WHOQOL-BREF scale. Descriptive analyses were carried out on the quantitative variables and Pearson's chi-square or McNemar's test on the categorical variables. Poisson regression was used in the cross-sectional study to check for associations between independent variables and low QoL. In the cohort, logistic regression was performed to verify the association between independent variables and worsening QoL. The cross-sectional study found that living in the north and east of Natal-RN and being born female significantly increased the likelihood of low QoL. Those with low (PR = 2.51; 95%CI 1.59-3.96) and medium (PR = 1.58; 95%CI 1.07-2.35) adherence to the Food Guide were also more likely to have low QoL. In the cohort, there was no significant difference (p>0.05) in QoL and in the frequency of categories of adherence to the Food Guide between pre- and post-pandemic, but there was a significant difference (increase in the frequency of agreement and decrease in disagreement) in the adoption of undesirable dietary practices (predominantly those involving consumption of ultra-processed foods) between the two moments of the study. After the pandemic, there was a tendency for people with per capita income in the 1st tercile and 2nd tercile to be more likely to have a worse overall QoL (adjusted OR = 0.02; p = 0.003) and (adjusted OR = 0.02; p = 0.004), respectively. In the cross-sectional study, it was concluded that adults and elderly people with low adherence to the Guide have a higher prevalence of low QoL, and in the cohort, the population started to adopt eating habits that can lead to health problems, in addition to per capita income having a significant association with reducing the likelihood of worsening QoL after the pandemic. Therefore, identifying the factors that determine its nutritional status is essential, ranging from food consumption to sanitary, environmental, socioeconomic, metabolic, and genetic aspects, which may increase the risk of vitamin E deficiency. In this context, this study evaluated the relationship of two factors with vitamin E nutritional status: 1) the dietary pattern, assessed through a systematic review (SR) that investigated the relationship between the consumption of ultraprocessed foods (UPF) and micronutrient status, including serum alpha-tocopherol; and 2) the COVID-19 pandemic, through the BRAZUCA-PÓS-COVID-19 cohort, which aimed to assess the impact of COVID-19 on vitamin E status in adults and older adults in Natal-RN. The SR followed the Preferred Reporting Items for Systematic Reviews and MetaAnalyses, with a protocol registered in PROSPERO (CRD42023488818). The search was conducted in the PubMed/MEDLINE, LILACS/SciELO, EMBASE, Scopus, and Web of Science databases, including human studies that analyzed nutritional biomarkers and the relationship between UPF and micronutrients. The cohort study involved 30 adult and elderly participants, of both genders, who had their serum vitamin E concentrations measured before and after the pandemic, in addition to socioeconomic, anthropometric, and biochemical data. The serum concentration of alpha-tocopherol was analyzed by Ultra High-Performance Liquid Chromatography. Vitamin E deficiency (VED) was considered with values ⦤ 12 μmol/L, and inadequacy < 30 μmol/L, corrected by the Total Cholesterol value. Of the three eligible studies in the SR, two were conducted to examine the relationship between UPF consumption and vitamin E status in pregnant women and lactating women. It was found that a higher amount of UPF in the diet was associated with a higher concentration of gamma-tocopherol in pregnant women (β=9.0, 95% CI 6.2, 11.8) and a lower concentration of serum alpha-tocopherol in lactating women (β= –0.17, 95% CI= −0.047, −0.010). In the cohort population, a reduction in serum alpha-tocopherol levels was observed post-pandemic (from 17.7 to 10.5 μmol/L), with an increase in VED cases from 45.8% to 54.2% (p<0.05). When alpha-tocopherol was adjusted for Total Cholesterol (μmol/L/mg), the median was 0.08 and 0.51 before and after the pandemic, respectively, showing a statistically significant negative change (Δ = -0.43; p<0.001). The results indicate negative outcomes from UPF consumption and the COVID-19 pandemic on vitamin E nutritional status, suggesting that dietary patterns and the public health emergency condition are factors influencing VED in this population. FCS management requires rigorous nutritional therapy with severe lipid restriction (less than 20 g per day) that can directly impact the intake and absorption of fat-soluble vitamins and cobalamin. This study aimed to evaluate the nutritional status of fat-soluble vitamins and cobalamin in individuals with FCS compared to healthy individuals. A case-control study matched by sex, age, and BMI category (1:1) was conducted, including 11 FCS patients matched with their respective controls (n=11), with participants recruited from Onofre Lopes University Hospital (HUOL) and the Federal University of Rio Grande do Norte (UFRN). Individuals with FCS were grouped according to genotypic alteration, being classified as heterozygotes or homozygotes. Data on lifestyle, health, physical examination, gastrointestinal symptoms, anthropometric assessment, and dietary intake were collected. Blood samples were obtained for biochemical analysis of the following vitamins: serum retinol, serum phylloquinone, serum alpha-tocopherol, 25-hydroxyvitamin D, and serum cobalamin. Data were analyzed using SPSS Statistics v.26.0 software, adopting a 95% significance level (p < 0.05). Effect size was calculated using Cohen's d, Cliff's Delta, Eta squared, and Epsilon squared. A large effect size was observed for retinol, with the heterozygote group showing higher levels compared to the homozygote and control groups. There was also a large effect size for alpha-tocopherol, where heterozygote and homozygote groups presented higher levels than the control group, the latter showing possible insufficiency of this vitamin according to the adopted cutoff points. Additionally, an inversely proportional correlation was identified between vitamin D and triglyceride, total cholesterol, and VLDL levels, as well as a directly proportional correlation between cobalamin and serum retinol. Regarding food consumption, it was verified that individuals with FCS had lipid intake above the recommended nutritional guidance of 20g/day. Thus, individuals with FCS presented significantly elevated concentrations of alpha-tocopherol and serum retinol compared to controls, with heterozygotes showing even higher retinol levels. Dietary lipid consumption exceeded nutritional recommendations by more than twice, suggesting an inadequate dietary pattern that may contribute to the syndrome's metabolic alterations.
