    Caracterização física e avaliação in vivo de um scaffold de hidroxiapatita/nióbio

    Hydroxyapatite (Hap) is a highly trained compound used for clinical use, due to the treatment of a mineral component present in the formation of bones, its use provides conditions for bone growth, thus allowing the biological fixation of grafts that are widely used for reconstructions of organic tissues, very little progress has been made in relation to materials capable of fulfilling with excellence the function of a material that is capable of having an osseointegration characteristic, in this sense the search for new constituent elements of the grafted pieces appears as a motivation of studies, as it provides the patient with an option to choose their treatments. In this sense, a Hydroxyapatite/Niobium compound was considered as a bioceramic material, it was characterized in terms of its morphology [X-ray diffraction (XRD), tension, cracking and tenacity and analyzed in vivo in a skullcap model with 8 mm critical defect performed in Wistar rats, with 3 experimental groups: Control (GC) - (positive control); Hap (hydroxyapatite standard) and (CNb/CPO - Hydroxyapatite/Niobio). After 90 days, the animals were euthanized and the samples were evaluated using Micro CT, HE and immunohistochemistry. The data obtained were statistically evaluated using the ANOVA test and the results were: In the mechanical results of XRD, the presence of hexagonal crystalline Hap and a P63/m space group was observed in the pure sample, while in the group with Nb, the presence of β-tricalcium phosphate (Ca3(PO4)2) and Fersmite (CaNb2O6), which may be related to the decomposition of Hap in the presence of Nb. In the XRF analysis, results close to those of grinding were obtained, proving that the technique used was efficient. Regarding the presence of indentations and cracks, there was a small increase in fracture toughness from Hap (0.78 MPa.m1/2) to Hap/Nb (0.82 MPa.m1/2). As in vivo results: micro-CT showed an increase in trabecular number (p˂0.001), a decrease in porosities (%) (p˂0.001) and a decrease in trabecular separation (mm) (p˂0.001) in the CNb/CPO group in comparison to the CG group. In the CNb/CPO group, there was the presence of new bone in which the connective tissue differentiates to form or indicate a bone matrix score of 3 (3-3.75, p˂0.05) and strong immunoexpression of osteopontin (p<0, 05). The results indicate that CNb/CPO showed resistance to crack propagation and stimulated bone formation in calvarial defects in rats.

    Perfil imunofenotípico das doenças linfoproliferativas crônicas no Rio Grande do Norte

    Chronic Lymphoproliferative Diseases (CLLD) constitute a group of lymphatic neoplasms characterized by the proliferation of mature neoplastic lymphocytes. Flow Cytometry Immunophenotyping (ICF) plays a fundamental role in the differential diagnosis of these disorders, contributing significantly to more accurate diagnostic accuracy. The main objective of this study was to demonstrate the immunophenotypic profiles used in the diagnosis of patients with CLBP in the state of Rio Grande do Norte (RN), evaluate the effectiveness of a specific panel for the diagnosis of Multiple Myeloma, determine the monoclonality of B lineage CLLD and establish a database for future clinical research. To achieve these objectives, we analyzed patients (n = 927) with CLLD using Flow Cytometry. Furthermore, we collected additional information from patients, such as age, sex and hematological parameters. The results obtained revealed that 85.7% of the cases corresponded to B lineage DLPC, while 14.2% were T/NK lineage DLPC. Among B-lineage CLLD, the distribution was as follows: 487 cases of Chronic Lymphocytic Leukemia, 18 of B Prolymphocytic Leukemia, 25 of Villous Cell Leukemia, 6 of Follicular Lymphoma, 4 of Splenic Lymphoma with Villous Lymphocytes, 33 of Mantle Cells, 4 from Waldenstron Macroglobulinemia, 69 from Multiple Myeloma, 14 from Plasma Cell Leukemia, 11 from Burkitt Lymphoma and 117 from Leukemized Non-Hodgkin Lymphoma. On the other hand, the classification of T/NK lineage CLLD included: 12 cases of Large Granular Lymphocyte Leukemia, 14 of T Prolymphocytic Leukemia, 18 of Adult T Cell Leukemia, 20 of Sézary Syndrome and 24 of Peripheral T Lymphoma The results demonstrate that Immunophenotyping by Flow Cytometry is a reliable and safe technique to clarify the diagnosis of patients with CLLD. Furthermore, it was possible to identify monoclonality in Blineage CLLD, and the use of a specific panel proved beneficial in the diagnosis of Multiple Myeloma.

    Phytol, a diterpene with notable effects on the central nervous system, has demonstrated anxiolytic properties in Swiss mice subjected to the elevated plus maze. Despite these findings, limited research has explored the impact of phytol nanoemulsions in animal models of anxiety, particularly in females. The present study aims to evaluate the effects of phytol incorporated into a sunflower oil nanoemulsion on rats exposed to the elevated plus maze. The formulation was developed using the phase inversion method, varying phytol concentration within the oil phase. The most stable formulation, containing the highest diterpene concentration, underwent extensive physicochemical characterization, including droplet size, polydispersity index (PDI), zeta potential, and pH over 35 days. Characterization techniques included fourier transform infrared spectroscopy, transmission electron microscopy, and chromatographic analysis. Additionally, the formulation was tested for hemolytic activity and antioxidant capacity using the DPPH assay and evaluated for its biocompatibility and delivery to the brain using a chicken embryo model. In vivo studies were performed in Wistar rats, with the animals divided into treatment groups (vehicle; diazepam 2 mg/kg; phytol 25 mg/kg; phytol 75 mg/kg; blank nanoemulsion; phytol nanoemulsion 25 mg/kg; and phytol nanoemulsion 75 mg/kg) administered via intraperitoneal injection. The physicochemical analysis revealed that the highest achievable phytol concentration in the nanoemulsion was 2.5%, with an average droplet size of approximately 160 nm, a PDI of 0.235, and a negative zeta potential of -27 mV, showing stability for over 30 days. Behaviorally, a significant reduction in anxiety-like behaviors was observed, as evidenced by increased entries and time spent in the open arms of the maze for rats treated with both the blank and phytol nanoemulsions at varying concentrations, compared to the vehicle group and the free phytol treatment at 25 mg/kg. These findings suggest that the phytol nanoemulsion formulation exhibits superior anxiolytic efficacy compared to the free phytol, highlighting the potential therapeutic advantage of the nanoemulsion system.

    The use of podocyte proteins as urinary biomarkers may be a promising noninvasive tool to detect glomerular injury at an early stage. After podocyte injury, the appearance of podocyte proteins in urine may identify glomerular injury before the onset of albuminuria. Early detection may contribute to the adoption of timely treatment, delaying or reducing disease progression. The objective was to analyze the scientific evidence on the use of podocyte proteins as urinary biomarkers of glomerular injury at an early stage. A protocol was developed and published in PROSPERO (CRD42021265630). A systematic review was performed following the recommendations of the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA). The search was performed for articles published until April 2024 in the PubMed, Embase, Web of Science, Scopus and Google Scholar databases, without language restrictions. The clinical question was formulated according to the Population, Index Tests, Reference Test, Diagnostic (PIRD) strategy, and observational case-control studies were included according to the eligibility criteria. Study selection was performed independently by three authors who extracted data using a standardized form developed and validated by the authors. Life-threatening risk was assessed using the Newcastle-Ottawa Scale (NOS). The R CORE TEAM, 2020 software was used for quantitative synthesis. The search strategy retrieved 3,476 studies, of which 20 were read in the integration phase and 8 were included in the review. The Grading of Recommendations Assessment, Development, and Evaluation (GRADE) method was used. Two studies presented low risk of bias and the other six presented moderate risk. The certainty of evidence for early-stage glomerular injury was considered low for podocin and very low for nephrin and podocalyxin. In conclusion, the studies developed individually demonstrate the potential of the proteins nephrin, podocin and podocalyxin in the diagnosis of glomerular injury in normoalbuminuric groups, allowing identification in the early stage of the disease.

    Complexo de inclusão entre óleo de copaíba (Copaifera multijuga Hayne ) e HPQCD: desenvolvimento e caracterização para fins anti Trypanosoma cruzi

    Chagas Disease, caused by the parasite Trypanosoma cruzi and primarily transmitted by the insect known as the 'kissing bug', represents a significant public health challenge in Latin America, affecting millions of people. In addition to medical implications, the disease is inherently linked to socio-economic and environmental factors such as disordered migrations, rapid urbanization, and environmental degradation. Given this challenging context, the search for new therapeutic strategies is crucial. In this context, this research aimed to develop and characterize inclusion complexes to improve the solubility of copaiba oil and evaluate its potential anti-Trypanosoma cruzi effect. For this purpose, copaiba oil and hydroxypropyl-G-cyclodextrin (OLC/HPGCD) were prepared by physical mixing (MF) and kneading methods (MLG). The complexes were then characterized using appropriate physicochemical methods. The results of the differential scanning calorimetry (DSC) showed endothermic peaks related to water loss, with HPGCD showing a peak at 70°C and OLC at 110°C, attributed to water molecule evaporation and volatilization, respectively. X-ray diffraction (XRD) analysis revealed crystallinity patterns characteristic of semicrystalline samples. Structural analyses by Fourier-transform infrared spectroscopy (FTIR) indicated the interaction between host and guest (OLC and HPGCD), demonstrating the potential of the developed inclusion complexes. The samples of HPGCD, MF, and MLG were analyzed with a scanning electron microscope (SEM) at various magnifications, revealing distinct morphological changes. HPGCD exhibited spherical particles, MF presented both spherical and fragmented particles, and MLG displayed irregular and porous shapes, indicating complete restructuring after processing.The anti-T. cruzi activities of HPGCD/OLC-MF and HPGCD/OLC-MLG complexes were comparable to Benznidazole at higher concentrations, although they were not effective at lower concentrations.

    People who are overweight or obese have more risk factors for developing cardiometabolic diseases. However, these factors present different degrees of cardiovascular risk depending on the metabolic profile. Considering the limitations and scarcity of specific assessment instruments and criteria in this context, the objectives of the present study are: 1) to develop and cross-validate two equations to estimate Fat Free Mass by bioimpedance analysis: one for the group of people with overweight or obesity (GSO) and another, for the group of people with obesity only (GO). 2) determine cut-off points for indices with anthropometric and biochemical parameters (IABP) for screening cardiometabolic risk factors (CMR) for the same population; 2) For objective 1, 305 people (134 women) were divided into two groups: development (n=206) and cross-validation (n=99); and for objective 2, the sample consisted of 330 subjects (192 women). For the study, DXA was used as a standard body composition technique, the tetrapolar monofrequency Bioelectrical Impedance Analysis (BIA) as equipment for using the equations, and the Visceral Adiposity Index (VAI), the Cardiometabolic Index (ICM) and the Triglycerides-Glucose Index (ITG) as IABP. To develop and validate the equations, the Paired t-test, multiple regression and Bland-Altman plot were used. To determine cutoff points, the ROC curve was plotted based on sensitivity and specificity, in addition to identifying the area under the AUC curve. Diagnostic agreement was calculated using Kappa analysis. The equation for GSO presented a correlation coefficient of agreement (CCC)=0.971; r2=0.94; standard error of estimate (SPE)=2.69kg; limits of agreement (LOA)=-4.4 to 4.3; and 2) and the equation for GO presented CCC=0.965; r2=0.93; EPE=2.27kg; LOA=-5.5 to 4.7. Women had the following cutoff points: IAV>4.11 (AUC=0.894), ITG>3.64 (AUC=0.858) and ICM>1.02 (AUC=0.912) and men: IAV>2.75 (AUC = 0.918), ITG>3.8 (AUC=0.888) and ICM>1.66 (AUC=0.923). When comparing the new models and the standard technique, there was no difference in the FFM value (p>0.05). The two derived equations are valid options for estimating the FFM of people with the characteristics of the studied sample, just as all IABP showed good results for screening RCM factors.

    Caracterização estrutural de compostos fenólicos de erythrina velutina willd. por espectrometria de massas associada às redes moleculares (molecular networking)

    The identification, characterization, and study of bioactive compounds can open doors to understanding numerous applications of interest for research. The arboreal species Erythrina velutina Willd., popularly known as “mulungu,” belonging to the Fabaceae family and the Papilionoideae subfamily, is distributed throughout the Brazilian Northeast. Its leaves, seeds, and mainly trunk bark are used in folk medicine in teas to treat sleep disorders and central nervous system (CNS) conditions. Previous works with the species relate its pharmacological, anxiolytic, and sedative properties, mainly to the presence of flavonoids and benzylisoquinoline alkaloids. Given the above, the study proposal aimed to characterize the phenolic compounds present in the leaves and seeds of the species collected in four different locations in the Seridó region of Rio Grande do Norte. Dereplication of the crude ethanol extract of seeds and leaves from different populations occurred with the aid of the GNPS (Global Natural Product Social Molecular Networking) platform, based on mass data and fragmentation patterns obtained by analysis by liquid chromatography coupled with mass spectrometry (LC-MS), which led to the formation of a molecular network that presented clusters attributed to phenolic compounds. A total of 136 compounds were annotated, being 32 flavonoids, two coumarins, two anthocyanins, an aurone, and a chalcone annotated by the platform library. The analysis of unannotated metabolites indicated similarities in the fragmentation pattern related to the aglycones present in the sample with some variations present in the substituents. Principal component analysis (PCA) for the substances noted, and present in samples from different populations of E. velutina made it possible to evidence which compounds allow the differentiation of the studied organs, seeds and leaves. In addition, the PCA analysis for the seed samples allowed to evidence the separation of the samples according to the collection region. Complementary methodologies were also developed and standardized for the analysis of metabolites by gas chromatography, which allowed the identification of different fatty acids such as palmitic, linoleic, oleic and stearic acids present in Erythrina seeds, through the esterification reaction, as well as other substances, such as phytosterols, silylated fatty acids and alkaloids, by silylation reaction with a derivatizing agent BSTFA. Thus, this work contributes to a greater understanding of the profile of phenolic compounds present in E. velutina and standardizing techniques that may contribute to the knowledge of the metabolome of this species in the future.

    The study of plants is under increasing transformation due to the availability of molecular and bioinformatics tools to understand metabolism and point out potential sources of new bioactive chemical entities with pharmaceutical interest. In this scenario, Erythrina velutina stands out as a medicinal plant found in the northeast region of Brazil, mainly in the Caatinga biome. This plant has a remarkable ability to produce alkaloids and flavonoids, which are the main secondary metabolites of the genus and are often associated with pharmacological activities, such as modulators of the central nervous system (with antidepressant and anxiolytic effects) and anti-inflammatory action. Although some natural products (NP) have been effective as a source of new drugs, the pharmaceutical industry still faces challenges in the identification and optimized production of these compounds and this has led to a reduction in interest in NP-based drugs. In order to minimize these gaps, this study aims to integrate molecular and chemical data to investigate the biosynthesis of the main metabolites present in E. velutina under three aspects: to evaluate the metabolic production in plants collected in natural habitat (Caatinga) and in cultivated plants submitted to different elicitors (ii); identify reference genes that normalize RT-qPCR tests for the species (ii) and evaluate the expression of target genes responsible for the production of alkaloids and flavonoids with bioactive potential (iii). The study focuses on transcriptome, proteome and metabolome data obtained from samples collected in their natural habitat, in addition to seedling cultivation subjected to environmental stresses (temperature, UV, water stress, salt stress and mechanical damage) and phytohormones (methyljasmonate , salicylic acid, nitric oxide and abscisic acid). As a result, the predominant flavonoids observed were flavones, isoflavones, and flavonols, along with several isoquinoline alkaloids from both dienoids and alkenoids. Transcriptome analysis revealed the presence of enzymes involved in the flavonoid and alkaloid biosynthesis pathways, including those differentially expressed, such as EvCHI, EvIF7GT, EvCHS, EvCOR, EvNMT and EvODM. Among the treatments tested, the most significant accumulation of metabolites occurred in response to NO, MeJA, water stress and heat, while ABA caused the least pronounced effect. For normalization of RT-qPCR assays, EvCSTF64, EvRAB7, EvNLE, EvUPF and EvARP4 were identified as the most stable reference genes, while EvNADH and EvPBL27 exhibited lower stability. These genes serve as reliable reference points to ensure accurate normalization of gene expression data in subsequent analyses. The results pointed to NO as crucial in increasing the expression of CHI and the amounts of isoflavones noted. Drought stress negatively affected early precursors of the Erythrina alkaloid pathway, while MeJA had the opposite effect. Exposure to heat increased alkaloid concentrations. This study will contribute to the understanding of the secondary metabolism of plants in the semi-arid/xeric environmental conditions, characteristic of the Caatinga region. In addition, may help to improve the production of bioactive molecules in the species and to value a plant from the Caatinga of Rio Grande do Norte as a source of compounds with pharmaceutical potential. This research represents the first step towards guiding the management for optimal target metabolite production in E. velutina.

    An important neglected tropical condition, the Chagas disease, though endemic to the Americas, has become a global health concern. The challenges in therapeutics highlight the urgency of developing vaccines to control this disease. Preclinical studies have expanded into a range of vaccine platforms and formulations to be tested. The presentation of new formulations using chimeric recombinant antigens of the etiological agent offers a consequent improvement in immunogenicity. In this context, this study focused mainly on the applicability of formulations with chimeric recombinant proteins of the infectious agent of Chagas disease, the protozoan Trypanosoma cruzi, used as antigenic candidates in isolation or in association with immunoadjuvant molecules and the characterization of the humoral immune response in different immunization protocols. For this purpose, the immunoadjuvant capacity of three structural polypeptides of the virus Triatoma virus (VP1+VP2+VP3: VPs) was presented and investigated, as well as vaccine formulations with the antigens with chimeric characteristics (IBMP-8.1, IBMP- 8.2, IBMP-8.3 and IBMP-8.4: IBMPs) in an antigenic mixture (Mix) associated or not with VPs and known adjuvants (Freund's Incomplete Adjuvant - FIA and Aluminum Hydroxide - Alum) through murine humoral immunity at different times during immunization protocols involving two doses (Protocol 1) and three doses (Protocol 2) in animal models. Thus, female Balb/c mice (Mus musculus) were immunized and in Protocol 1 were separated into five groups: Mix, Mix+FIA, Mix+Alum, Mix+VPs and negative control, while in Protocol 2 the groups were: FIA, Alum, Mix, Mix+FIA, Mix+Alum, and negative control. An experimental infection protocol with T. cruzi was also presented. Animals from all protocols had specific murine antibody profiles of the total IgG and its subtypes: IgG1, IgG2a, IgG2b, and IgG3 determined by Enzyme-Linked Immunosorbent Assay (ELISA) from serum samples. The data demonstrate that the VPs were able to induce good levels of total IgG, IgG2a, IgG2b and IgG3 antibodies and no induction of IgG1, thus characterizing the humoral immunostimulatory property of these polypeptides in association with IBMPs antigens in Mix form; they also suggest that Mix directs to a profile of low presence or total absence of IgG1, an interesting possibility for immune responses against intracellular agents. Additionally, they show that aluminum hydroxide presents performance similar at different concentrations and, finally, they offer the development of two immunization protocols in a murine model in the context of Chagas disease, as well as the importance of the booster dose. Moreover, immunized animals recognize the parasite and infected animals recognize the IBMPs proteins. This pioneering work in the panorama of chimeric recombinant antigens of T. cruzi associated with VPs, FIA and the most used adjuvant in vaccines (Alum), presents itself as promising for the development of new immunobiologicals. The findings promoted the optimization of immunization protocols with small amounts of antigens capable of inducing the production of total IgG antibodies and subtypes that persist after booster doses and provided valuable insights to drive the advancement of chimeric vaccines for human infections caused by trypanosomatids, such as Chagas disease.
