Artigo Evaluation of medicines advertising in medical journals(Universidade de São Paulo (Faculdade de Ciências Farmacêuticas), 2009-06) Souza, Daliana Maria Berenice de Oliveira; Lima, Suellen Cristiane Medeiros de; Batista, Almária Mariz; Carvalho, Maria Cleide Ribeiro Dantas deEste trabalho objetivou analisar propagandas de medicamentos sujeitos à prescrição, que foram veiculadas em três periódicos das áreas de cardiologia e neurologia, dirigidos a profissionais de saúde. A análise envolveu critérios baseados na legislação em vigor, além de outros, e das referências bibliográficas. Averiguou-se a presença dos seguintes itens: número de registro, nome do fármaco, indicações terapêuticas, contra-indicações, cuidados e advertências, reações adversas, efeitos colaterais, posologia, legibilidade das informações técnico-científicas e referências bibliográficas, frases e/ou expressões sobre vantagens ante outros fármacos, mensagens de segurança, promessas de cura e imagens de pessoas sorridentes, além da confirmação das citações com base nas referências bibliográficas. Dentre os critérios legais avaliados, observou-se a ausência de legibilidade das informações técnico-científicas em 85% das peças promocionais; a ausência dos efeitos colaterais em 23%; a ausência de cuidados e advertências em 15%; de contra-indicações em 12,8%; de posologia em 6,4%; do número de registro em 2,7% e da Denominação Comum Brasileira/Denominação Comum Internacional (DCB/DCI) em 0,6%. Das 130 afirmações relativas a benefícios frente a outros fármacos, 23,8% não foram confirmadas; e das 48 mensagens de segurança veiculadas, 41,7% não tinham respaldo nas referências citadas. As imagens de pessoas sorridentes foram recurso usado em 49,2 % das peças promocionais. Neste sentido, dispositivos legais foram criados para assegurar o uso desses produtos, sem que haja interferência no aleitamento materno. Este estudo objetivou analisar propagandas de alimentos abrangidos pela Lei nº 11.265/2006, que regulamenta a comercialização e publicidade de alimentos para lactentes e crianças de primeira infância no Brasil, no período de junho 2006 a maio de 2008, na cidade de Natal/RN. Foram coletadas 220 propagandas, sendo 141 de material técnico-científico e 79 de promoção comercial que foram avaliadas quanto às exigências legais. O resultado desta análise demonstrou que 100% das propagandas veiculadas em material técnico-científico infringiram o inciso V; 18,4% o inciso IV e 14,2% os incisos I, II e III do artigo 19 da Lei nº 11.265/2006. Em 42% das promoções comerciais não foram veiculadas as informações obrigatórias constantes dos incisos I e II do artigo 5º. The objective of this study was to detect and identify the products of enzymatic hydrolysis of chitosan (dimers to hexamers) using a crude extract of chitosanolytic enzymes produced by the fungus Metarhizium anisopliae. These fungus was able to produce, during 48 h cultivation in a medium containing chitosan, chitooligosaccharides ranging from dimers, trimers, tetramers and pentamers at concentrations 0.2, 0.19, 0.06, 0.04 mg/mL, respectively, and the enzymatic activity was 2.5 U/L. Using the crude enzyme extract for chitosan hydrolysis, we detected the presence of dimers to hexamers at hydrolysis times of 10, 20, 30, 40, 50 and 60 min of enzymatic reaction, but the yields were higher at 10 min (54%). Escherichia coli has been the predominant prokaryote used in genetic engineering studies due to the abundance of information about its metabolism. Despite significant advances in molecular biology and immunology of infections, there are as yet no prophylactic drugs capable of preventing visceral leishmaniasis. It is therefore important to identify specific antigens in order to develop vaccines and diagnostic kits against this disease. The objective of this study was to evaluate the influence of culture medium on the production of eIF antigen from Leishmania chagasi in recombinant Escherichia coli. In this work, we examined the cytotoxicity of pure COS and oligomers A, B and C (solutions composed of different amounts of COS) produced by enzymatic hydrolysis using a crude enzyme extract produced by the fungus Metarhrizium anisopliae. The antiproliferative effect of these molecules was analyzed using tumor cell lines (HepG2 and HeLa cells) and in a normal cell line (3T3). The antioxidant activity was analyzed in several in vitro experiments. Glucosamine showed higher toxicity (approximately 92%) to all cell lines studied. However, the oligomers obtained after hydrolysis demonstrated no toxic effects on the normal cells (3T3). Furthermore, we showed that a small amount of other COS can decrease the cytotoxic effect of glucosamine against 3T3 cells, indicating that glucosamine could be used as an antitumor drug in the presence of other COS. In addition, different effects were found in antiproliferative assays, which depended on the COS composition in the oligomers (A, B and C), showing that a combination of them may be essential for developing antineoplastic drugs. Superoxide anion scavenging was the main antioxidant activity demonstrated by the COS and oligomers. This activity was also dependent on the oligomer composition of the chitosan hydrolysates. Further work will identify the ideal proportions of COS and glucosamine for maximizing the effects of these biological activitiesArtigo Avaliação da propaganda de medicamentos veiculada em emissoras de rádio(ABRASCO - Associação Brasileira de Saúde Coletiva, 2013-02) Batista, Almária Mariz; Carvalho, Maria Cleide Ribeiro Dantas deO objetivo deste estudo foi avaliar propagandas de medicamentos veiculadas em emissoras de rádio em Natal (RN), Brasil, nos períodos de abril a setembro de 2008 e abril a setembro de 2010. As propagandas foram captadas, gravadas e transcritas para se efetuar uma análise legal e uma de conteúdo baseada nos preceitos de Laurence Bardin. Tanto as captadas no primeiro período (sob égide da RDC 102/00), quanto às do segundo (sob égide da RDC 96/08), apresentaram algum tipo de infração legal. A análise de conteúdo detectou, em ambos os períodos, praticamente os mesmos vícios como ausência de informações negativas sobre o medicamento, apelo ao consumo, exaltação da eficiência/eficácia e exploração abusiva de enfermidades. Apesar da mudança para uma legislação mais moderna e mais restritiva, as propagandas de medicamentos, no rádio, continuam a infringir frontalmente a legislação, cometendo abusos e desrespeitando o direito à saúde da população. Here, we evaluated the production potential of biologically functional chitooligosaccharides using enzymes extracts produced by Paenibacillus chitinolyticus and Paenibacillus ehimensis. After 48 h of fermentation, these microorganisms were able to produce chitosanases, which generated oligomers with a degree of polymerization between dimers and hexamers. The maximum conversion of chitosan to oligomers was 99.2 %, achieved after 12 h incubation of chitosan with enzymes produced by P. ehimensis. The chitooligosaccharides generated were capable of scavenging the 2,2-diphenyl-1-picrylhydrazyl radical, reaching a maximum scavenging rate of 61 and 39 % when produced with P. ehimensis and P. chitinolyticus enzymes, respectively. No ensaio em batelada, o IPTG foi adicionado ao cultivo na concentração final de 1 mM. Na batelada alimentada, 0,2 mM do indutor foi adicionado a cada hora, totalizando uma concentração final de 0,8 mM. O procedimento de indução por pulsos de IPTG indicou um incremento de aproximadamente 49% na produção de biomassa quando comparado em batelada. Pode-se concluir que a estratégia de indução empregando pulsos de IPTG (batelada alimentada) foi mais eficaz na expressão do antígeno 503 quando comparado à estratégia de batelada simples. The main available specific treatment is the antivenom serum therapy, which has some disadvantages, such as poor neutralization of local effects, risk of immunological reactions, high cost and difficult access in some regions. In this context, the search for alternative therapies is relevant. Therefore, the aim of this study was to evaluate the antiophidic properties of Jatropha gossypiifolia, a medicinal plant used in folk medicine to treat snakebites. The aqueous leaf extract of the plant was prepared by decoction and phytochemical analysis revealed the presence of sugars, alkaloids, flavonoids, tannins, terpenes and/or steroids and proteins. The extract was able to inhibit enzymatic and biologic activities induced by Bothrops jararaca snake venom in vitro and in vivo. The blood incoagulability was efficiently inhibited by the extract by oral route. The hemorrhagic and edematogenic local effects were also inhibited, the former by up to 56% and the latter by 100%, in animals treated with extract by oral and intraperitoneal routes, respectively. The inhibition of myotoxic action of B. jararaca reached almost 100%. According to enzymatic tests performed, it is possible to suggest that the antiophidic activity may be due an inhibitory action upon snake venom metalloproteinases (SVMPs) and/or serine proteinases (SVSPs), including fibrinogenolytic enzymes, clotting factors activators and thrombin like enzymes (SVTLEs), as well upon catalytically inactive phospholipases A2 (Lys49 PLA2). Antiinflammatory activity, at least partially, could also be related to the inhibition of local effects. Additionally, protein precipitating and antioxidant activities may also be important features contributing to the activity presented. (Euphorbiaceae) is a medicinal plant largely used in folk medicine. Teas from the leaves are popularly used as an antithrombotic agent and the branches are frequently employed as a “thick blood” agent. Considering that the anticoagulant activity associated with antioxidant properties could be beneficial for various cardiovascular diseases, this study’s aim is the evaluation of anticoagulant and antioxidant activities of J. gossypiifolia leaves, seeking new therapeutic purposes for this plant. Methods: The aqueous leaf crude extract (CE) was prepared by decoction and was fractionated by liquid-liquid partition with solvents of increasing polarity. The phytochemical analysis was performed by thin layer chromatography (TLC) and by the spectrophotometric quantification of sugars, proteins and phenolic compounds. The anticoagulant activity was evaluated by prothrombin time (PT) and activated partial thromboplastin time (aPTT) tests. The capacity to act in the fibrinolytic system (fibrinolytic and fibrinogenolytic activities) was also assessed. The antioxidant activity was evaluated by total antioxidant capacity, reducing power, copper chelating activity, iron chelating activity, hydroxyl radical scavenging activity and superoxide radical scavenging assays. The potential toxicity was evaluated using hemolytic assay and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide (MTT) assay on HEK-293 cells. Results: CE showed significant anticoagulant activity in aPTT test, while no action was observed in PT test, suggesting a preferential action toward the intrinsic and/or common pathway of coagulation. No effect was observed in the fibrinolytic system. Using the aPTT test, it was observed that the residual aqueous (RA) fraction was the most active, being two times more active than CE. RA presented very significant antioxidant activity in all models tested comparable to or even higher than CE. Regarding the safety, CE and RA did not produce significant cytotoxicity in both tests employed. Phytochemical analysis revealed the presence of alkaloids, flavonoids, proteins, tannins, steroids and/or terpenoids and sugars. Anti-leishmanial treatment is problematic because antileishmanial drugs are toxic and costly and drug resistance is emerging. Moreover, there is no effective and safe vaccine approved for human use against any form of leishmaniasis. It seems likely that both a subunit vaccine and an optimized diagnostic test would benefit from the production of recombinant antigens. However, for recombinant protein production, the composition of the cell growth medium must be carefully formulated and monitored, because it may have significant metabolic effects on both the cells and protein expression. Thus, the aim of this study was to optimize the culture medium for cell growth and the expression of the 648 antigen from Leishmania i. chagasi in recombinant Escherichia coli M15. The induction allowed the expression of the 648 antigen; this was confirmed by electrophoresis, which showed a protein with a molecular mass of 24 kDa. The results showed that the higher cell concentration obtained in TB medium can be explained by the larger number of components such as amino acids and vitamin precursors contained in this complex medium, as well as glycerol as a carbon and energy source. Acetic acid excretion, detected at the end of fermentation, resulted in an inhibitory effect for both cell growth and protein production, because this was above the concentration limit of 0.9 g L−1Artigo Armazenamento de Medicamentos em Unidades Básicas de Saúde de Macaíba, RN, Brasil(Conselho Federal de Farmácia - CFF, 2016-03-31) Simão, Renato Wagner Tinôco de Sousa; Batista, Almária MarizEste estudo avaliou o armazenamento de medicamentos em 22 unidades básicas de saúde do município de Macaíba, Estado do Rio Grande do Norte, no período de janeiro a março de 2012. Para isto, foi realizada verificação in loco da infraestrutura e observação da rotina do serviço destas unidades, além de aplicação de Roteiro de Inspeção, com base na RDC nº 44/09, o que proporcionou subsídios para contextualização do papel do farmacêutico frente à assistência farmacêutica no local. Para identificação e análise dos medicamentos com prazo de validade expirado, foram utilizadas a Relação Nacional de Medicamentos Essenciais e a Relação Municipal de Medicamentos Essenciais do município. Nenhuma das farmácias destas unidades apresentava responsável técnico farmacêutico, manual sobre armazenamento de medicamentos, controle de umidade e temperatura. Em todas as farmácias havia medicamentos com prazo de validade expirado, abrangendo 1395 unidades mínimas não fracionadas, pertencendo a 31 classes farmacológicas diferentes e abrangendo 34 especialidades farmacêuticas. Tomou por base a tradução e adaptação dos indicadores validados no Projeto ISEP-Espanha, além do documento Safe Practices for Better Healthcare do National Quality Forum dos Estados Unidos, que possui 34 recomendações de boas práticas. Realizou-se validação por um painel de 25 especialistas e análise da confiabilidade e viabilidade em um estudo-piloto realizado em três hospitais com diferentes tipos de gestão (estadual, federal e privada). Aprovaram-se 75 indicadores de boas práticas (39 de estrutura; 36 de processo) para 31 das 34 recomendações. The effects of pH, ammonium sulfate concentration, OPP concentration, air flow-rate, flotation time and initial volume of organic phase were evaluated on process performance using multivariate statistical methodology. Under optimal conditions, the separation efficiency (E%), the concentration coefficient (a) and the distribution coefficient (D) for ATPF reached 57.55%, 4.23 and 9.86, respectively. When compared to others conventional techniques also used in this work, such as aqueous two-phase extraction, solvent sublation and liquid-liquid extraction, the ATPF showed better performance in terms of E%, a and D. In this study, we prepared three B. trimera extracts aqueous extract (AE), decoction (AE-D), and methanol extract (ME) and investigated their antioxidant effects in six different tests and their anti-adipogenic effect in 3T3-L1 cells. The extracts showed a dose-dependent antioxidant activity in all tests. AE was the most potent antioxidant in copper and ferric ion chelation assays, whereas AE-D was the most potent in superoxide and hydroxyl radical scavenging assays, reducing power assay, and total antioxidant capacity analysis. Only ME showed a cytotoxic effect against 3T3-L1 cells. Lipid accumulation decreased in 3T3-L1 adipocytes in the presence of AE and AE-D extracts (0.5 to 1.0 mg/mL). In addition, the extracts dramatically attenuated the levels of adipogenic transcriptional factors, including CCAAT enhancer-binding protein α (C/EBPα), CCAAT enhancer-binding protein β (C/EBPβ), and gamma receptors by peroxisome proliferators (PPARγ), during adipogenesis. AE-D (1.0 mg/mL) caused an approximately 90% reduction in the levels of these molecules. The natural and pretreated biomass was characterized using the National Renewable Energy Laboratory protocol analysis as well as X-ray diffraction and scanning electron microscopy. The results demonstrated that in terms of the total reducing sugars, there was no significant difference between the hydrolysis using Triton X-100 and rhamnolipids, regardless of the pretreatment. A cellulosic conversion value as high as 33.0% was obtained in experiments with rhamnolipids. The coconut husk was observed to be a potential biomass that could produce second generation ethanol, and the rhamnolipid preparation can be used to support for the enzymatic hydrolysis, enhancing the advantage of cellulose conversion into glucose over chemical surfactants because it is an environmentally friendly approach
