    Scorpion bites are responsible for significant morbidity and pediatric mortality in many parts of the world and represents a public health problem in Brazil. Tityus stigmurus is a scorpion specie of medical importance that occur in the Brazilian Northeast. In this context, this work proposed to analized the inflammatory and pathophysiological parameters after Tityus stigmurus scorpion envenoming in vivo by intraperitoneal injection of 375 µg/kg of venom in BALB/c mice to evaluate its local and systemic effects. For this purpose, the total and differential leukocyte number, myeloperoxidase activity, nitrite levels and total protein were quantified in peritoneal exsudate, as well as, serum biochemical parameters and cytokine (TNF-α and IL-10) levels were analyzed. Alterations in relative organs weight of the experimental animals were also assessed. The results showed that the venom was not able to induce the migration of cells into the peritoneum of experimental animals. However, significant biochemical abnormalities were observed as the increase in CK, CK-MB, AST, ALT, creatinine, amylase, and uric acid. Moreover, it was observed lesions in organs such as heart, liver, kidney and pancreas. It was also demonstrated edema response that was characterized by rapid onset, lasting up to 1 hour. This is the first time that partial characterization of the envenoming induced by T. stigmurus in mice was carried out.

    Amphotericin B is a water-insoluble compound widely used to effectively treat systemic fungal infections. The use of more effective therapies with higher doses of AmB directed to invasive systemic mycoses, together with a real need to reduce AmB intrinsic toxicity, has trigged the development of alternative carriers for AmB administration. Lipid microemulsions have been very important in the pharmaceutical field, once they are pharmacologically more efficient and present reduced side effects. Thus, this work aimed to evaluate the stability and the pharmacological and the toxic profile of AmB incorporated in a microemulsion system (AmB-ME), and to compare its performance with free and conventional AmB. To assess AmB stability, the formulations were subjected to a stress study (thermal stress and UV luminous radiation) and accelerated stability studies for 90 days, in which the formulations were stored at 25ºC and 45ºC. Then, at pre-determined times, the AmB content, followed by activity and toxicity assays, was evaluated. The results showed that AmB has higher susceptibility to UV luminous radiation rather than high temperatures. For all formulations, AmB presented a first order kinetic degradation profile. However, AmB-ME revealed the shortest degradation profile for a shelf-life of almost 30 days. Regarding the antifungal activity, the AmB-ME formulation was able to inhibit C.parapsilosis growth during 90 days of the experiment. Nevertheless, AmB-ME showed higher toxicity levels, as revealed by potassium and hemoglobin release assays. Thus, the microemulsion can be considered a very interesting strategy to stabilize AmB under the stress conditions, but it is necessary to optimize the product formulation to minimize possible cellular damage.

    The plasmids (pDNAs) have been standing out regarding to its application at biotechnology, mostly in DNA vaccination and genic therapy. pDNAs are DNA molecules with extra chromosomal double bands, presenting circular shape and have different isoforms. The Food and Drug Administration recommends that during pDNAs utilization on immunization protocols, they must be at least 80% on its super-curled isoform, once the studies assign that super-curled isoforms have a larger access to the interior of cells. Super curled isoform may be altered by chemical, physical and biological factors. Owing to its importance at pDNAs structure stability at biological function, this study aims to evaluate the pDNAs stability used at biotechnology, ands tests a nanoformulation with polyethyleneimine (PEI) associated to pDNAs, in order to promote a stability and bioavailability increasing of pDNAs. We evaluate the temperature effect at incubation, plasmid size and nucleases concentrations under super-curled isoform conservation. Also, it was evaluated pDNAs stability using functionalized nanoparticles with PEI through time variables, nucleases concentration and comparing to non functionalized nanoparticles formulated pDNAs. On 37°C and 42°C temperatures, there was higher degradation on plasmids, as well as higher serum concentration, presenting super-curly isoforms only at 1:64 dilution. The smaller sized plasmid presented lower degradation in all the tests. The preparation of functionalized nanoparticles with PEI associated to pDNA showed to be a promising alternative of protection against enzymatic degradation, with 100% efficiency of plasmid association. However, it requires more in vitro and in vivo studies to verify security factors and genic transference rate.

    Accidents caused by snakes are considered a major public health problem in several regions of the world. Bothrops are responsible for the vast majority of ophidic accidents in Brazil. It is known that, currently, the only specific treatment available for the bite of these snakes is the bothropic antivenom, but it has some limitations, such as not combat local effects effectively, difficult access in some regions, and high cost. Plants of the genus Jatropha (Euphorbiaceae) are often indicated by traditional medicine, in some regions of Brazil, to reverse the damage caused by snake bites and also with anti-inflammatory function. In this view, the objective of this work was to evaluate the anti-inflammatory, healing and antiophidic potential against the venom of the snake Bothrops jararaca of the hydroethanolic extract of the leaves of Jatropha mollissima (Pohl) Baill. incorporated into a formulation developed for topical application. Tests were performed to evaluate the topical anti-inflammatory activity of this species in carrageenan-induced paw edema and ear edema induced by single application (acute inflammation) or multiple applications (chronic inflammation) of chroton oil. Tests were also performed to evaluate the healing activity of the formulation, while the skin irritation test was conducted to analyze safety during topical use of the formulations developed. It was possible to observe that the gel had anti-inflammatory action in acute inflammation models induced by carrageenan, and in acute and chronic inflammation models by chroton oil. chroton oil. It was also observed that the gel has cicatrizant activity, since it significantly decreased the size of the wound in relation to the placebo group. The gel did not cause skin irritation in mice, proving to be safe for topical use. The efficacy of the gel containing the hydroethanolic extract of the leaves of J. mollissima alone and in association with the commercial polyvalent antibothropic-crotalic commercial antivenom in relation to its antiophidic potential against the local effects of B. jararaca envenoming were evaluated in the paw edema and muscle hemorrhagic models and it was seen that the venom-induced edematogenic and hemorrhagic activities were inhibited, both by the gel alone and by the association with the antiophidic antivenom, since these treatments reduced edema (the gel treatment containing the extract in association with the antiophidic antivenom had an inhibition of approximately 90% after four hours of the envenoming), in addition to significantly reducing the concentration of hemoglobin in the evaluated tissues, consequently having a significant anti-hemorrhagic potential. In conclusion, the results show that the gel containing the hydroethanolic extract of the leaves of J. mollissima shows potential anti-inflammatory, healing and antiophidic activity, acting on the local effects of the bite, suggesting that this formulation of topical use has an important antiophidic potential as a complementary alternative to the treatment of local effects caused by bothropic envenoming, besides acting as healing potential and topical antiinflammatory for the treatment of various skin diseases.

    As atividades antiinflamatória e antinociceptiva foram estudadas para o extrato seco padronizado de Phyllanthus niruri L., (PnSDE) em modelos experimentais de inflamação e dor. A atividade antiinflamatória do PnSDE foi avaliada por meio dos modelos de edema de pata induzido por carragenina, migração leucocitária induzida por tioglicolato e artrite induzida por zymosan. A atividade antinociceptiva foi avaliada utilizando os testes de Randall e Selitto, retirada da calda e placa quente. Este estudo mostrou que o extrato seco padronizado de Phyllanthus niruri L (PnSDE) apresentou importante atividade antiinflamatória frente aos modelos experimentais de edema de pata, migração leucocitária e artrite induzida por zymosan. Assim, o PnSDE, nas doses utilizadas reduziu de forma significativa a resposta vascular no processo inflamatório induzido por carragenina e inibiu a migração leucocitária para o local da inflamação induzida por tioglicolato. Em relação ao modelo de artrite, o PnSDE conseguiu reduzir de maneira significativa os níveis da enzima mieloperoxidase e provocar uma elevação dos níveis de glutationa total. Neste modelo, a análise histopatológica das sinóvias revelou uma significativa redução dos eventos inflamatórios avaliados no tecido inflamado. Na avaliação imunohistoquímica das sinóvias, os resultados sugerem que o extrato pode ter contribuído com a redução da expressão para RANKL, RANK, OPG, COX-2, MMP-2 e MMP-9. Na avaliação da atividade antinociceptiva, os ensaios realizados evidenciaram que o PnSDE exibiu uma marcante atividade antinociceptiva periférica e central. Em conclusão, este estudo sugere que o extrato seco padronizado de Phyllanthus niruri L. possui potentes atividades antiinflamatória e antinociceptiva. Este estudo contribui para uma possível obtenção de uma preparação fitoterápica, utilizando o extrato seco padronizado de Phyllanthus niruri (PnSDE), validando o seu uso para o tratamento da dor e distúrbios inflamatórios.

    Tuberculosis is a disease of older, known in the worid. The main treatment for this disease is achieved with the fixed-dose combination formulations, involving the main drugs. This study aims to determine the quality and conduct the first evaluation of stability of capsules of isoniazid and rifampicin formulations nationals with respect to physical, Chemical properties and dissolution after storage in accelerated conditions (40 ° C temperature and 75% RH) and under conditions of long duration (30 ° C temperature and 75% relative humidity). USP methods were used previously validated, based on visible spectrophotometry for the dissolution of rifampicin and high performance liquid chromatography for the content of rifampicin and isoniazid and disbandment. Four samples were submitted to six months of accelerated stability study and 12 months of stability study of long duration. The drugs were subjected to stressful conditions, performing hydrolysis at different pH values and temperatures. All formulations tested showed satisfactory for pharmacopoeial testing. In degradation studies, there were products resulting from the conditions imposed on drugs. But a more careful analysis, using methods more sensitive and specific instruments is needed to separate and quantify these degradation products. The formulations tested are nationals of good quality and monitoring the appearance of degradation products is needed throughout the stability study.

    Accidents with venomous animals, mainly snakes and scorpions, are considered a serious public health problem. In Brazil, ophidian accidents are mainly provoked by snakes of the genus Bothrops, and those with scorpions, by the species Tityus serrulatus. The conventional treatment is the use of antivenom serotherapy, which in addition to presenting some limitations, such as difficult access in some regions and high cost, is only able to neutralize the poison toxins, being inefficient in inhibiting the endogenous reactions triggered by their presence. In this context, the aim of this paper was to perform a phytochemical analysis of the fruit juice of Hancornia speciosa and its ability to inhibit the effects of the venom of the scorpion Tityus serrulatus and the snake Bothrops jararaca. The phytochemical analysis was performed using high performance liquid chromatography MS/MS. The antiscorpionic activity of the juice was evaluated using the animal model of acute pulmonary edema induced by the scorpion venom T serrulatus. For this, Swiss mice received venom subcutaneously and were treated orally with juice at doses of 100 and 200 mg / kg, using pre- and post-treatment protocols. Two hours after venom administration, the animals were euthanized to collect lungs, kidneys and blood. The following analyzes were performed with the lungs: cytokine quantification, evans blue concentration, myeloperoxidase enzyme (MPO) dosage and relative lung weight percentage; with the kidneys, nitrite dosage and lipid peroxidation; and with blood, hematological and biochemical parameters were evaluated. Regarding botropic venom, the ability of the juice to inhibit its local effects was evaluated in the models of paw edema, hemorrhage, myotoxicity and dermonecrosis. For this, Swiss mice of both sexes were used. In all models the juice was administered orally at doses of 100 and 200 mg / kg and was administered shortly after the botropic venom (post-treatment protocol). Through HPLC-DAD-MS / MS analyzes 13 compounds were identified, the major ones identified as L-bornesitol, quinic acid, chlorogenic acid and rutin. Observations regarding the inhibition of antiscorpionic effects demonstrated that Hancornia speciosa juice was able to significantly decrease some important parameters such as cytokine production, myeloperoxidase activity, evans blue concentration in lung tissue, lipid peroxidation and nitrite. Regarding the botropic effects, the juice was able to significantly reduce the dermonecrotic lesion, the edematogenic activity, the hemorrhagic activity and the concentrations of the creatine kinase and lactate dehydrogenase enzymes. From the results, it can be concluded that Hancornia speciosa Gomes juice has great potential to be used as adjuvant in the treatment of scorpion and botropic poisonings.

    The Berardinelli-Seip syndrome (SBS) is a rare autosomal recessive disease characterized by a near complete absence of adipose tissue and metabolic disorders such as hypertriglyceridemia, hyperlipidemia and diabetes mellitus. The reduction of serum leptin concentration is another important finding in the SBS and is related to changes in adipocyte function. Leptin is an adipocytokine that plays an important role in the regulation of various systems, including the immune system, acting directly or indirectly on immunity. This study aimed to identify the presence of immunological abnormalities and evaluate the relationship between changes found and leptin deficiency. The study was approved by the Ethics Committee and were included 28 subjects, 10 males and 18 females, all diagnosed with SBS. Analyses were performed biochemical serum and leptin, in addition to evaluation of hematologic parameters, immunoglobulin dosages (IgA, IgG, IgM and IgE), complement proteins (C3 and C4) and the count of lymphocyte subpopulations (T lymphocytes, B and natural killer cells) using immunophenotyping by flow cytometry. All study subjects had significantly lower leptin averaging 1.00±0,45ng/mL. Immunological evaluation showed an increase in the concentrations of immunoglobulins and complement proteins compared with serum leptin was no statistically significant difference in IgA, IgM, C3 and C4. In the assessment of immunophenotype was no statistically significant difference with B lymphocyte count (CD19+) (p<0,05) compared with leptin. The results showed alterations in innate and adaptive immune response. The changes found may involve the development of various diseases, contributing mainly to the increased risk of infections. Additional studies are needed to confirm and assist in the investigation of the effect of leptin on immunity.

    O DM esta relacionada a diversas complicações, entre elas a osteopenia/osteoporose. Oobjetivo deste estudo foi avaliar o efeito de suplementação com minerais e antioxidantes nadieta associado ao tratamento com insulina, sobre o metabolismo ósseo em modelo deosteopenia diabética por um período de 60 dias. Foram utilizados ratos Wistar machosdivididos em 4 grupos: Grupo C - Controle (n=5), Grupo D - Diabéticos experimentais porSTZ (n=6), Grupo DI - Diabéticos experimentais por STZ tratado com insulina (n=6) e GrupoDSI - Diabéticos experimentais por STZ suplementados com 2,5 vezes de cálcio e fósforo,20X de vitamina E e 500mg/Kg de dieta de zinco, e tratados com insulina (n=7). Foramavaliados parâmetros bioquímicos como a atividade séricas da fosfatase alcalina total (ALP),concentrações séricas de cálcio total, cálcio ionizável, fósforo e glicose, bem como análisehistomorfométrica através da espessura trabecular óssea, volume ósseo e distanciatrabecular e parâmetros moleculares através da análise da expressão gênica do mRNA dosgenes RANK/RANKL/OPG. Foram considerados diabéticos os animais que apresentaraglicemia 250 mg/dl. Os animais diabéticos apresentaram um estado de hiperglicemiaassociado ao quadro de poliúria, polidpsia, polifagia e a perda de peso corporal. Observouseuma aumento nas concentrações séricas de cálcio total nos grupos D, DI e DSI quandocomprado com o grupo C (p=0,0393; plt;0,0001 e plt;0,0001 respectivamente), bem como umaumento na concentração sérica do íon nos grupos DI e DSI quando comparado com ogrupo D (p=0,0017 e p=0,0063 respectivamente). Paras as concentrações séricas de cálcioionizável observou-se uma diminuição da concentração sérica de Ca ionizável nos grupos D,DI e DSI quando comparado com o grupo C (plt;0,032, plt;0,0001 e pgt;0,0001respectivamente). O fósforo sérico não apresentou diferenças significativas. A atividade daenzima ALP total mostrou-se aumentada no grupo D, DI e DSI comparado com o grupo C(p=0,0083, p=0,0015, p=0,0265 respectivamente). A distancia trabecular mostrou-seaumentada para o grupo D comparado ao controle (plt;0,001) e diminuída nos grupos DI eDSI comparado ao D (plt;0,001). A espessura trabecular óssea apresentou-se diminuída nogrupo D (p=0,0065) comparado ao controle e aumentada nos grupos DI e DSI comparadoao D (p=0,0065; p=0,0088 respectivamente). O volume ósseo apresentou valoresdiminuídos no grupo D comparado aos grupos DI e DSI (p=0,0006; p=0,0264respectivamente). A expressão do mRNA do gene RANK apresentou uma diminuição naexpressão nos grupos D, DI e DSI quando comparado com o grupo C (p=0,0122, plt;0,0001e plt;0,0001 respectivamente), e uma diminuição da expressão nos grupos DI e DSI emrelação ao grupo D (plt;0,0001). Para o mRNA do gene RANKL não observa-se diferençassignificativas entre os grupos. A expressão do mRNA do gene OPG mostrou uma diminuiçãono grupo D, DI e DSI quando comparado com o grupo C (p=0,0495, plt;0,0001 e plt;0,0001respectivamente). O aumento da distância trabecular e a redução do volume e espessuratrabecular óssea apresentado pelo grupo diabético sugerem a possibilidade de perda demassa óssea associada com o estado hiperglicêmico e o aumento da espessura e volumetrabecular óssea associado a redução da distancia trabecular, bem como redução naexpressão dos genes RANK e OPG e aumento da relação RANKL/OPG dos grupos tratadoscom insulina, sugerem um efeito anabólico da insulina, no qual reflete em uma redução deperda de massa óssea.

    Susceptibilidade do Triatoma brasiliensis (Hemiptera: Reduviidae Triatominae) à infecção por isolados do Trypanosoma cruzi (Kinetoplastida, Trypanosomatidae) do Estado do Rio Grande do Norte

    The susceptibility of Triatoma brasiliensis to infection by Trypanosoma cruzi DTUs I, II and III found in the State of Rio Grande do Norte was evaluated experimentally. Nymphs were fed using direct and indirect xenodiagnosis with the isolates 3188 (TcI), RN79/RN2 (TcII) and Pl1.10.14 (TcIII) in simple or mixed infections. The intestinal contents were evaluated by direct exam, xenoculture and kDNA polymerase chain reaction (PCR). Then, the characterization of isolates was used the cytochrome oxidade subunit 2 gene (COII), 24Sα ribosomal DNA gene (rDNA) and the spliced-leader intergenic region of the mini-exon gene (SL-IR) as markers. In the direct xenodiagnosis, the direct exam presented 30.0%(15/50) of positivity, the xenoculture 18.0%(9/50) and the kDNA PCR 88.0% (44/50). Meanwhile, the indirect xenodiagnosis showed 5.4%(3/56) of positivity to the direct exam, 25.0%(14/56) to xenodiagnosis and 41.1%(23/56) to kDNA PCR. Comparing the techniques, the direct xenodiagnosis showed better results than the indirect procedure. The PCR identified the T. cruzi in both types of infection, with higher positivity than direct exam and xenoculture. With respect the molecular characterization, the re-isolates showed the same profile before and after the simple infections. In the mixed infections were verified two or only one isolate. The data demonstrated the susceptibility of T. brasiliensis to DTUs I, II and III, with high rates in simple and mixed infections reinforcing the importance of T. brasiliensis to the epidemiology of T. cruzi infection in Rio Grande do Norte.
