Familial hypercholesterolemia (FH) is an autosomal dominant disease of genetic origin that directly increases levels of low-density lipoprotein (LDL-c). Due to its LDL-c alteration profile, FH increases the chances of developing atherosclerotic diseases, such as atherosclerotic heart disease (ICD-10 I.25.1) and atherosclerotic cardiovascular disease (ICD-10 I.25.0). Consequently, the primary treatment for FH is commonly the use of a maximum statin dosage. Utilizing a set of RNA-Seq public data from a study on the use of statins in patients with FH and an epidemiological dataset on ICD-10 I25.1 deaths, our objective is to carry out an RNA-Seq data analysis, integrating with information described in the literature and correlated epidemiological insights, in order to bring new perspectives that have not yet been proposed. A thorough analysis of epidemiological data obtained from public databases revealed that 2017 was the year with the highest number of ICD-10 I.25.1 death notifications, while 2020 and 2021 were the years with the lowest number of notifications. Furthermore, age and sex analyses indicated higher male mortality and an increase in deaths with age, with notable reductions among individuals over 80 during the pandemic. Finally, the analysis of RNA-Seq data revealed 15 altered genes in the ”before and after” comparison of statin use, as determined by concatenating the results of the three methods used (DGE, DTE, and DTU). Of these 15 altered genes, we identified two (LMBRD2 and HFE) that suggested new anti-atherosclerotic mechanisms promoted by statinspt_BR
