(Desenvolvimento e Meio Ambiente, 2021-05-27) Mota, Ana Carolina Costa Campos; Gomes,Ana Francisca Teixeira; Porciúncula, Laura Brito; Chaves, Viviany Moura; Almeida, Adriana Monteiro de; Jorge, Thiago Perez; Jacob, Michelle Cristine Medeiros; https://orcid.org/0000-0001-5503-0096; https://orcid.org/0000-0003-1265-0089; https://orcid.org/0000-0002-9654-7152; https://orcid.org/0000-0001-5447-8593; https://orcid.org/0000-0002-0375-8613; https://orcid.org/0000-0002-4881-7285

Background: The Pharmaceutical industry sponsorship, research outcome and quality has been already evaluated for clinical trials in order to analyze if this kind of sponsorship affects the results of clinical trials. In this sense, this study has the aim to investigate whether placebo use allows positive outcomes regarding efficacy and safety compared to synthetic medicines. Methods: We designed and registered a study protocol for a systematic review for methodology data. We will only randomized clinical trials that use placebo as comparator. The main outcome will be the evaluation of placebo use regarding the tendency for positive results (efficacy and security) when comparing to synthetic medicines. PubMed, Cochrane, LILACS (BVS), Web of Science, Scopus, and Excerpta Medica dataBASE (EMBASE) databases will be searched. Gray literature will be identified through the databases Proquest (Dissertation and Theses), OpenGrey and Google Scholar. Two review authors will independently assess trial quality and will extract data in accordance with standard Cochrane methodology. If necessary, we will also contact authors for additional information. The Cochrane Collaboration’s risk of bias tool will be used. If feasible, it means homogenous data, we will conduct random effects meta-analysis. Subgroup analyses will be conducted for different justifications for placebo use and for studies sponsored/not sponsored by the pharmaceutical industry. Results: Our present findings will indicate the effects of placebo use as comparator regarding efficacy and safety of the oral synthetic medicines. Discussion: This systematic review will identify, summarize, and analyze if there is a trend for positive efficacy and safety results for synthetic medicines in clinical trials when compared with placebo and if the justification for placebo use is considered ethically acceptable. Validation of the QualiPresc instrument for assessing the quality of drug prescription writing in primary health care
(Plos One, 2022) Souza, Dyego Leandro Bezerra de; Batista, Almaria Mariz; Gama, Zenewton Andre da Silva; https://orcid.org/0000-0001-8426-3120
Introduction: Adverse events related to drug prescriptions are the main patient safety issue in primary care; however there is a lack of validated instruments for assessing the quality of prescription writing, which covers the prescriber, the patient and the drug information. Objective: To develop and validate the QualiPresc instrument to assess and monitor the quality of drug prescriptions in primary care, accompanied by a self-instruction direction, with the goal of filling the gap in validated instruments to assess the quality of prescription writing. Methodology: A validation study conducted in a municipality in Northeastern Brazil, based on prescriptions prepared in January 2021 by physicians assigned to 18 Basic Health Units and filed in 6 distribution/dispensing units. Four steps were covered: 1) Analysis of content validity of each indicator (relevance and adequacy); 2) Analysis of reliability via intra and inter-rater agreement of each indicator; 3) Analysis of the utility of each indicator; 4) Construction and analysis of the reliability of a weighted composite indicator based on effectiveness and safety scores for each indicator. Results: Twenty-nine potential indicators were listed, but only 13 were approved for validity, reliability and usefulness. Twelve indicators were excluded because of validity (<90% validity index) and four because they were not useful in the context of the study. Three weighted composite indicators were tested, but only one was approved for reliability and usefulness. The validated instrument therefore contains 13 indicators and 1 weighted composite indicator. Conclusion: This study demonstrates the validity, reliability and usefulness of QualiPresc for the evaluation of prescription writing in the context of primary care. Application to contexts such as secondary care and tertiary care requires cross-cultural adaptation and new content validity. Educators, managers and health care professionals can access QualiPresc online, free of charge, to assess performance and provide feedback involving drug prescribers

Quality of prescription writing in Brazilian primary health care
(Primary Health Care Research & Development, 2023) Souza, Dyego Leandro Bezerra de; Batista, Almária Mariz; Gama, Zenewton André da Silva; Hernández, Pedro Jesús Saturno; https://orcid.org/0000-0001-8426-3120
Objective: To evaluate the quality of prescription writing in the context of public primary health care. Background: Prescription errors are one of the leading patient safety problems in primary care and can be caused by errors in therapeutic decisions or in the quality of prescription writing. Methods: Cross-sectional observational study conducted in a municipality in Northeastern Brazil. The assessment instrument (including 13 indicators and one composite indicator) was applied to a representative sample of drug prescriptions from the 24 Family Health Teams providing Primary Health Care in the municipality, dispensed in January 2021. Estimates of compliance and their 95% confidence intervals and graphical analysis of frequencies are assessed globally and stratified by dispensing units and prescribers. Findings: The average composite prescription writing quality on a 0-100 scale was 60.2 (95% CI 57.8–62.6). No quality criteria had 100% compliance. The highest compliance rates were found for ‘frequency of administration’ (98.9%) and ‘identification of the prescriber’ (98.9%). On the other hand, ‘recorded information on allergy’ (0.0%), ‘patient’s date of birth’ (1.7%), ‘nonpharmacological recommendations’ (1.7%), and ‘guidance on the use of the drug’ (25%) were the indicators with lower compliance, contributing to 69% of the noncompliances found. The type and frequency of the errors in the quality of prescription writing uncovered in this study confirm the continuing need to tackle this problem to improve patient safety. The results identify priority aspects for interventions and further studies on the quality of prescription writing in the context of Primary Health Care in Brazil

Natural latex serum: characterization and biocompatibility assessment using galleria mellonella as an alternative in vivo model
(Taylor and Francis, 2021-12-30) Brasil, Giovana Sant’Ana Pegorin; Barros, Patrícia Pimentel de; Miranda, Matheus Carlos Romeiro; Barros, Natan Roberto de; Junqueira, Juliana Campos; Gomez, Alejandro; Herculano, Rondinelli Donizetti; Mendonça, Ricardo José de; https://orcid.org/0000-0003-4885-2811
Natural latex serum (NLS) is one of the natural rubber latex fractions from Hevea brasiliensis tree, which is formed by centrifuged serum and is composed of proteins, acids, nucleotides, salts and carbohydrates. The proteins present in NLS have demonstrated several interesting biological properties, including angiogenic, healing, osteogenic, anti-inflammatory, antimicrobial, in addition to inducing neovascularization, bone formation and osseointegra tion. Thus, we proposed to characterize NLS by physicochemical techniques and to investigate the biocompatibility by toxicological assays and safety test in Galleria mellonella. Infrared spectrum showed vibrational bands characteristic of amide I, II and III that are linked to the protein content, which was confirmed by the High Performance Liquid Chromatography profile and by the Electrophoresis analysis. This material did not exhibit hemolytic (rate <0.5%) and cytotoxic effects (viability >70%) and was able to enhance the proliferation of fibroblasts (>600%) after 3 days. The pronounced proliferative effect observed in fibroblast cells can be explained by the presence of the fibroblast growth factor (FGF) like protein revealed by the Western blot test. Moreover, NLS did not provoke toxic effects (survival ~ 80%) on the G. mellonella model, indicating that it is a biocompatible and safe material

(Frontiers Media, 2021-08-02) Barros, Patrícia Pimentel de; Rossoni, Rodnei Dennis; Terra-Garcia, Maíra; Kaminski, Valéria de Lima; Loures, Flávio Vieira; Fuchs, Beth Burgwyn; Mylonakis, Eleftherios; Junqueira, Juliana Campos; https://orcid.org/0000-0003-4885-2811
Candida albicans is the main fungal species associated with the development of oral candidiasis. Currently, therapeutic options for these infections are limited by the adverse effects of antifungal drugs and by the emergence of drug resistant strains. Thus, the development of new antifungal agents is needed for the prevention and treatment of oral Candida infections. Caffeic acid phenethyl ester (CAPE) is a natural compound from propolis polyphenolic groups that exhibits many pharmacological properties. In this study, we investigated whether CAPE can have antifungal and immunomodulatory effects on oral candidiasis. Preliminary tests to assess the antifungal activity of CAPE were performed using the Minimum Inhibitory Concentration (MIC) assay that demonstrated inhibition in a range from 16 to 32 mg/mL, confirming its antifungal activity on several C. albicans strains isolated from the oral cavity. Subsequently, we analyzed Candida spp biofilms formed in vitro, in which CAPE treatment at 5 x MIC caused a reduction of 68.5% in the total biomass and ~2.60 Log in the viable cell count (CFU/mL) in relation to the untreated biofilm (p<0.0001). Next, RNA was extracted from untreated and CAPE-treated biofilms and analyzed by real-time qPCR. A series of genes analyzed (ALS1, ECE1, EPA1, HWP1, YWP1, BCR1, BGR1, CPH1, EFG1, NDT80, ROB1, TEC1, UME6, SAP2, SAP5, PBL2, and LIP9) were downregulated by CAPE compared to the untreated control group (p<0.0001). In in vivo studies using Galleria mellonella, the treatment with CAPE prolonged survival of larvae infected by C. albicans by 44.5% (p < 0.05) and accompanied by a 2.07-fold increase in the number of hemocytes. Flow cytometry revealed the most prominent increases were in types P2 and P3 hemocytes, granular cells, which phagocytize pathogens. In addition, CAPE treatment decreased the fungal load in the hemolymph and stimulated the expression of antifungal peptide genes such as galiomicin and gallerimycin. The antifungal and immunomodulatory activities observed in G. mellonella were extended to a murine model of oral candidiasis, in which CAPE decreased the levels of C. albicans colonization (~2 log CFU/mL) in relation to the untreated control group. In addition, CAPE treatment significantly reduced pseudomembranous lesions, invasion of hyphae on epithelium surfaces, tissue damage and inflammatory infiltrate (p < 0.05). CAPE was also able to increase the expression of b-defensin 3 compared to the infected and untreated group by 3.91-fold (p < 0.0001). Taken together, these results show that CAPE has both antifungal and immunomodulatory effects, making it a promising natural antifungal agent for the treatment and prevention of candidiasis and shows impact to oral candidiasis

(Elsevier, 2021-06) Terra-Garcia, Maíra; Souza, Cheyenne Marçal de; Gonçalves, Nathalia Maria Ferreira; Pereira, André Henrique Correia; Barros, Patrícia Pimentel de; Borges, Alessandra Bühler; Miyakawa, Walter; Strixino, Juliana Ferreira; Junqueira, Juliana Campos; https://orcid.org/0000-0003-4885-2811
Photodynamic therapy (PDT) is a promising strategy to control cariogenic pathogens, such as Streptococcus mutans. Seeking to reach the total bacterial elimination from dental surfaces, novel photosensitizers have been investigated, such as Fotoenticine (FTC) derived from chlorin e6. The objective of this study was to investigate the photodynamic effects of FTC against several clinical strains of S. mutans. Clinical isolates were obtained from patients with active carious lesions, identified by molecular analysis and subjected to PDT using laser irradiation (660 nm and 39.5 J/cm2) in planktonic and biofilm stages. We identified 11 S. mutans strains from cervical, occlusal and proximal caries. PDT mediated by FTC has totally eliminated the S. mutans cells in planktonic growth for all analyzed strains. In biofilms, PDT with FTC reached statistically significant reductions compared with the non-treated control group, at 5.4, 5.5 and 6.5 Log10 (CFU/mL), respectively, for the strains from proximal, occlusal and cervical caries. The scanning electron microscopy evaluations confirmed that PDT mediated by FTC was able to disaggregate and kill the S. mutans cells adhered to enamel surface, suggesting its potential to disinfect the dental tissues

Objective: To analyze the self-esteem of pregnant women admitted to the high-risk ward of a reference hospital in the city of Recife. Materials and Methods: This is a cross-sectional, descriptive study with a quantitative approach. The sample population included 112 pregnant women.Data collection was performed through a form adapted from the version in the Brazil of the Rosenberg Self-Esteem Scale (RSES), plus socioeconomic, obstetric and reproductive questions. The sample included pregnant women admitted to the high-risk sector, and excluded pregnant women under age and those who presented inability of understanding and/or verbalization to answer the form. The data analysis was performed by means of descriptive statistics and the results, presented as tables. Results: 72.3% had unsatisfactory self-esteem. Conclusion: The low self-esteem was elevated in high-risk pregnant women. There is need for updating/training health professionals from high-risk maternity hospitals to identify signs of symptoms of low self-esteem in this population with the purpose of offering a reception and a suitable care
